ABSTRACT The habenula has been implicated in psychiatric disorders such as depression, primarily because of its role the modulation dopaminergic and serotonergic systems, which play a pathophysiology these disorders. Despite growing evidence supporting behavioral regulation, process by neural cells develop remains elusive. Since habenular anlage is found prosomere 2 domain expressing transcription factor Dbx1 mouse embryos, we hypothesized that Dbx1‐expressing source astrocytes modulate neuronal activity habenula. To address this, examined cell lineage generated from male mice using tamoxifen‐inducible Cre recombinase under control promoter. Perinatal induction labeled migrating radially ventricular zone to pial side anlage, eventually showed astrocyte‐like morphology with expression marker protein, S100β, for mature adult mouse. Photostimulation ChR2 released potassium ions into extracellular space, turn excited neurons an increased firing rate lateral Finally, photostimulation exacerbated depression‐like phenotypes reduced locomotor activity, exaggerated despair behavior impaired sucrose preference open‐field, tail suspension tests, respectively. These results indicated perinatal modulated via regulation levels.
