The emergence and evolution of SARS-CoV-2 variants, such as Delta Omicron, pose significant challenges to pandemic management. This study evaluated the effectiveness reverse-transcription polymerase chain reaction (RT-PCR) whole-genome sequencing (WGS) in detecting characterizing variants using 624 samples collected South Korea from mid-2021 mid-2022. Two RT-PCR genotyping assays demonstrated a high concordance rate (90.4%) identifying variant during its dominance. In contrast, WGS revealed extensive genetic diversity among Omicron sub-lineages, 29 distinct including two Korea-specific (BA.1.1.5 BA.2.3.8). Clustering analysis data highlighted groupings BA.1, BA.2, BA.5 with overlap shared mutations suggesting evolutionary convergence. Sub-lineage expanded rapid transmission phases subsequently consolidated dominant lineages emerged. These findings highlight complementary strengths underscore importance integrating these methodologies for effective monitoring public health response.

With the aim of developing novel anti-SARS-CoV-2 drugs to address ongoing evolution and emergence drug-resistant strains, reported SARS-CoV-2 Mpro inhibitor WU-04 was selected as a lead find novel, highly potent, broad-spectrum inhibitors. Using fragment-based multilevel virtual screening strategy, 15 hit compounds were identified subsequently synthesized. Among them, A5 (IC50 = 1.05 μM), A6 1.08 A9 0.154 μM) demonstrated potent inhibition comparable or slightly weaker than WU-04. Antiviral activity evaluations revealed that compound exhibited strongest antiviral with an EC50 value 0.18 μM, quite marketed drug Nirmatrelvir (EC50 0.123 inferior 0.042 μM). Molecular dynamics simulations elucidated key interactions between A5, A6, A9, binding pocket Mpro, providing valuable insights into their mechanisms action. These findings identify promising for development.

Background/objectives: Attitudes towards COVID-19 vaccination vary globally, influenced by political and cultural factors. This research aimed to assess the views of people without a healthcare qualification in Europe on safety, effectiveness, necessity as well how informed they felt. The secondary outcomes focused respondents’ were affected demographic context factors included comparison country level feeling informed. Methods: A mixed-method cross-sectional online survey eight European countries, using convenience sampling. Results: total 1008 adults completed survey, 60% whom female. While only 44.1% considered vaccines safe, 43.5% effective, 44.9% necessary, 80.0% had been vaccinated. Four ten strongly agreed that informed, while over quarter did not answer question. Our previous study has found that monoclonal antibodies targeting a conserved epitope peptide spanning from residues 1144 to 1156 of SARS-CoV-2 spike (S) protein, namely S(1144-1156), can broadly neutralize all the prevalent strains, including wild type, Alpha, Epsilon, Delta, and Gamma variants. In study, S(1144-1156) was conjugated with bovine serum albumin (BSA) formulated Montanide ISA 51 adjuvant for inoculation in BALB/c mice its potential as vaccine candidate. Results showed titers S protein-specific IgGs neutralizing mouse sera against various variants, Omicron sublineages, were largely induced along three doses immunization. The significant release IFN-γ IL-2 also observed by ELISpot assays through stimulating vaccinated splenocytes peptide. Furthermore, vaccination S(1143-1157)- S(1142-1158)-EGFP fusion proteins elicit more than S(1144-1156)-EGFP protein. Interestingly, antisera collected inoculated exhibited better efficacy BA.2.86 JN.1 subvariants BA.1, BA.2, XBB subvariants.
