Abstract Marine molluscan cell lines, required for virus screening and cultivation, form essential tools developing health management strategies these animals in the blue economy. Moreover, they are also crucial to develop cultivated seafood. As there is no valid marine line, primary cultures relied upon all investigations. A sound protocol generating from molluscs entailed, but existing protocols often involve heavy antibiotic usage depuration that invariably affect gene expression health. This work presents an easy‐to‐adopt, time‐saving using non‐depurated mollusc Crassostrea madrasensis , which requires only initial treatment minimal exposure or use of antibiotics culture medium. The important experimental considerations arriving at this have been elucidated. Accordingly, sodium hypochlorite neomycin sulfate were chosen disinfecting tissues. study first shrimp medium (SCCM) as a culture. Despite being osmoconformers, oysters exhibited stable intracellular osmotic conditions pH, which, when provided vitro, promoted effective cardiomyocyte formation. viability could be enhanced 10% fetal bovine serum (FBS), healthy obtained SCCM without FBS. optimized allowed regular beating clusters retained month. The application of high-throughput sequencing technologies to non-model organisms has brought new opportunities for the identification bioactive peptides from genomes and transcriptomes. From this point view, marine invertebrates represent a potentially rich, yet largely unexplored resource de novo discovery due their adaptation diverse challenging habitats. Bioinformatics analyses available genomic transcriptomic data allowed us identify myticalins, novel family antimicrobial (AMPs) mussel Mytilus galloprovincialis, similar AMPs Modiolus spp., named modiocalins. Their coding sequence encompasses two conserved N-terminal (signal peptide) C-terminal (propeptide) regions hypervariable central cationic region corresponding mature peptide. Myticalins are taxonomically restricted Mytiloida they can be classified into four subfamilies. These subject considerable interindividual variability possibly presence/absence variation. Functional assays performed on selected members indicate remarkable tissue-specific expression (in gills) broad spectrum activity against both Gram-positive Gram-negative bacteria. Overall, we present first linear ever described in mussels confirm great potential bioinformatics tools organisms.

Abstract Background Oysters in coastal environments are subject to fluctuating environmental conditions that may impact the ecosystem services they provide. Oyster-associated microbiomes responsible for some of these services, particularly nutrient cycling benthic habitats. The effects climate change on host-associated microbiome composition well-known, but functional changes and how host physiology functioning poorly characterized. We investigated parameters affect oyster-associated microbial community structure function along a trophic gradient Narragansett Bay, Rhode Island, USA. Adult eastern oyster, Crassostrea virginica, gut seawater samples were collected at 5 sites this estuarine August 2017. Marine invertebrate microbiomes play important roles in diverse host and ecological processes. However, a mechanistic understanding of host-microbe interactions is currently available for small number model organisms. Here, an integrated taxonomic functional analysis the microbiome eastern oyster, Crassostrea virginica, was performed using 16S rRNA gene-based amplicon profiling, shotgun metagenomics, genome-scale metabolic reconstruction. Relatively high variability observed across individual oysters among different tissue types. Specifically, significantly higher alpha diversity inner shell than gut, gill, mantle, pallial fluid samples, distinct composition revealed gut compared to other tissues examined this study. Targeted metagenomic sequencing microbiota led further characterization dominant bacterial taxon, class Mollicutes, which captured by reconstruction metagenome-assembled genome (MAG). Bivalve mollusks thrive in environments rich microorganisms, such as estuarine and coastal waters, they tend to accumulate various particles, including viruses. However, the current knowledge on mollusk viruses is mainly centered few pathogenic viruses, whereas a general view of bivalve-associated viromes lacking. This study was designed explore viral abundance diversity bivalve using transcriptomic datasets. From analyzing RNA-seq data 58 species, we have reconstructed 26 nearly complete over 413 partial RNA virus genomes. Although 96.4% predicted proteins refer new some sequences belong associated with species or other marine invertebrates.
