The maintenance of epithelial barrier function involves cellular tension, with cells pulling on their neighbors to maintain integrity. Wounding interrupts which may serve as an early signal initiate repair. To characterize how wounds alter we used a laser-recoil assay map cortical tension around in the monolayer Drosophila pupal notum. Within minute wounding, there was widespread loss along both radial and tangential directions. This similar levels observed Rok inactivation. Tension subsequently restored wound, first distal then proximal cells, reaching wound margin about 10 minutes after wounding. Restoring required GPCR Mthl10 IP3 receptor, indicating importance this calcium signaling pathway known be activated by damage. restoration correlated inward-moving contractile wave that has been previously reported; however, itself not affected knockdown. These results indicate transiently increase contract absence signaling, but is critical for fully resetting baseline it disrupted

After a tissue is wounded, cells surrounding the wound adopt distinct wound-healing behaviors to repair tissue. Considerable effort has been spent on understanding signaling pathways that regulate immune and tissue-resident as they respond wounds, but these signals must ultimately originate from physical damage inflicted by wound. Tissue wounds comprise several types of cellular damage, recent work indicates different initiate signaling. Hence understand signaling, it important identify localize wound-induced damage. Laser ablation widely used researchers create reproducible, aseptic in can be live-imaged. Because laser wounding involves combination photochemical, photothermal photomechanical mechanisms, each with spatial dependencies, around pulsed-laser will experience gradient Here we exploit this map Using genetically-encoded fluorescent proteins, monitor damaged sub-cellular components epithelial living Drosophila pupae seconds minutes following wounding. We hypothesized regions would predictably arrayed varying sizes, subsequent analysis found all radii are linearly related over 3-fold range size. Thus, estimated after measuring any one. This report identifies within wounded animal. By quantitatively mapping size placement set foundation for tracing back initiates it.

Wound healing is an aspect of normal physiology that we all take for granted until it goes wrong, such as, example, the scarring results from a severe burn, or those patients who suffer debilitating chronic wounds fail to heal. Ever since wound repair research began as discipline, clinicians and basic scientists have collaborated try understand cell molecular mechanisms underpin healthy in hope this will reveal clues therapeutic treatment pathological healing. In recent decades mathematicians physicists begun join with important challenge. Here describe examples how mathematical modeling married biological experimentation has provided insights biology alone could not fathom. To date, these studies largely focused on re-epithelialization inflammation, but also discuss other components might be ripe similar interdisciplinary approaches.

Summary The clearance of dead and dying cells, termed efferocytosis, is a rapid efficient process one that critical for organismal health. extraordinary speed efficiency with which cells are detected engulfed by immune within tissues presents challenge to researchers who wish unravel this fascinating process, since these fleeting moments uptake almost impossible catch in vivo. Abstract Nutrient sensing and the subsequent metabolic responses are fundamental functions of animals, closely linked to diseases such as type 2 diabetes various obesity-related diseases. Drosophila melanogaster has emerged an excellent model for investigating metabolism its associated disorders. In this study, we used live-cell imaging demonstrate that fly functional homolog mammalian glucagon, Adipokinetic hormone (AKH), secreted from AKH hormone-producing cells (APCs) in corpora cardiaca, stimulates intracellular Ca 2+ waves larval fat body/adipose tissue promote lipid metabolism. Further, show specific dietary amino acids activate APCs, leading increased secretion. Finally, a comparison dynamics adult bodies revealed different mechanisms regulation, highlighting interplay pulses secretion, extracellular diffusion hormone, intercellular communication through gap junctions. Our study underscores suitability powerful exploring real-time nutrient inter-organ dynamics.

All organisms have evolved to respond injury. Cell behaviors like proliferation, migration, and invasion replace missing cells close wounds. However, the role of other wound-induced cell is not understood, including formation syncytia (multinucleated cells). Wound-induced epithelial were first reported around puncture wounds in post-mitotic Drosophila epidermal tissues, but more recently been mitotically competent tissues such as pupal epidermis zebrafish epicardium. The presence active suggests that offer adaptive benefits, it unknown what those benefits are. Here, we use vivo live imaging analyze pupae. We find almost half near a wound fuse form large syncytia. These several routes speed repair: they outpace diploid complete closure; reduce intercalation during pool resources their component concentrate them toward wound. In addition healing, these properties are likely contribute roles development pathology.

ABSTRACT All organisms have acquired mechanisms for repairing themselves after accidents or lucky escape from predators, but how analogous are these across phyla? Plants and animals distant relatives in the tree of life, both need to be able efficiently repair themselves, they will perish. Both an outer epidermal barrier layer a circulatory system that must protect infection. However, plant cells immotile with rigid cell walls, so cannot raise animal-like immune response move away insult, as can. Here, we discuss parallel strategies signalling pathways used by plants heal their tissues, well key differences. A more comprehensive understanding parallels differences could highlight potential avenues enhance healing patients’ wounds clinic and, reciprocal way, developing novel alternatives agricultural pesticides.
