ABSTRACT During aging, animals experience a decline in proteostasis activity, including loss of stress-response activation, culminating the accumulation misfolded proteins and toxic aggregates, which are causal onset some chronic diseases. Finding genetic pharmaceutical treatments that can increase organismal lengthen life is an ongoing goal current research. The regulation stress responses by cell non-autonomous mechanisms appears to be potent way impact healthspan. In this Review, we cover recent findings intersection with special focus on articles preprints published between November 2021 October 2022. A significant number papers during time increased our understanding how cells communicate each other proteotoxic stress. Finally, also draw attention emerging datasets explored generate new hypotheses explain age-related collapse.

C-Terminal cyclic imides are posttranslational modifications that can arise from spontaneous intramolecular cleavage of asparagine or glutamine residues resulting in a form irreversible protein damage. These damage events recognized and removed by the E3 ligase substrate adapter cereblon (CRBN), indicating these aging-related may require cellular quality control mechanisms to prevent deleterious effects. However, factors determine peptide susceptibility C-terminal imide formation their effect on stability have not been explored detail. ABSTRACT Skeletal muscle is a highly adaptable tissue, finely tuned by various physiological and pathological factors. Whilst the pivotal role of skeletal in overall health widely acknowledged, unravelling underlying molecular mechanisms poses ongoing challenges. Protein ubiquitylation, crucial post‐translational modification, involved regulating most biological processes. This widespread impact achieved through diverse set enzymes capable generating structurally functionally distinct ubiquitin modifications on proteins. The complexity protein ubiquitylation has presented significant challenges not only identifying ubiquitylated proteins but also characterising their functional significance. Mass spectrometry enables in‐depth analysis modification status, offering powerful tool for studying its diversity: an approach termed ubiquitylomics. Ubiquitylomics been employed to tackle different perspectives including limited global quantification substrates linkages, site recognition crosstalk with other modifications. As field mass continues evolve, usage ubiquitylomics unravelled novel insights into regulatory governing biology. However, research predominantly conducted cellular models, limiting our understanding signalling events driving By integrating intricate landscape dynamic shifts physiology, promises deepen biology lay foundation developing transformative muscle‐related therapeutics. review aims articulate how can be utilised researchers address aspects muscle. We explore methods used experiments, highlight relevant literature employing context outline considerations experimental design.

Since the first description of a set characteristics aging as so-called hallmarks or pillars in 2013/2014, these have served guideposts for research biology. They been examined range contexts, across tissues, response to disease conditions environmental factors, and benchmark various anti-aging interventions. While were intended capture generalizable aging, they are derived mostly from studies rodents humans. Comparative including species animal tree life great promise reveal new insights into mechanistic foundations there is diversity lifespan age-associated physiological changes. However, it unclear how well defined apply diverse species. Here, we review each twelve by Lopez-Otin 2023 with respect availability data We evaluate current methods used assess their potential be adapted comparative studies. Not unexpectedly, find that supporting described restricted humans few model systems no available many clades. Similarly, not all can easily assessed at least half hallmarks, today employed fill this gap knowledge, suggesting prioritized while developed study remaining hallmarks.
