In this review, it is evaluated the progress in application of stem cell therapy to ameliorate symptoms bipolar disorder, major depression, schizophrenia, and autism. These disorders are highly prevalent clinical medicine responsible for high levels psychosocial disability among patients. All them share common biomedical features, such as complex variable genetic substrates, significant susceptibility environmental changes, insufficient knowledge their pathogenesis. addition, responsiveness patients pharmacological treatment heterogeneous, some cases, no available. Therefore, development cell-based regenerative its possible combination with emerging therapeutic approaches that promote neural plasticity expected advance neuropsychiatry next few decades.

Abstract Embryonic stem cells (ESCs) can break symmetry and differentiate along different lineages, even when exposed to a seemingly identical environment. It is thought that this priming of towards lineages due cell-cell variation, although the underlying mechanisms are poorly understood. To address this, we exploit tractability social amoeba Dictyostelium discoideum, where cell fate choice also does not depend on spatial cues. We develop test model explain quantitative experimental single observations probabilistic differentiation. The suggests cycle position affects lineage choice, as previously shown, but stochastic variation plays key role. Single sequencing reveals genes strongly associated with exhibit extensive expression variation. Like in ESCs, they specific epigenetic modifications, which perturbed affect their disrupt choice. suggest represents an adaptive mechanism increases developmental robustness against perturbations deterministic signals.
