The concept of a 'magic bullet' was first introduced by Paul Ehrlich in the early 1900s, he foresaw advent targeted therapies and specific killing harmful cells and/or microorganisms. However, these were only used clinic after second half 20th century with development monoclonal antibodies. To date, 13 antibody-drug conjugates (ADCs) are commercially available. Many advances have been made modifying one or several three main components an ADC, namely antibody, cleavable non-cleavable linker payload, integrating conjugation chemistry. Despite efforts, some problems emerged thus limit their effectiveness. New strategies could overcome identify next generation ADC.

We report the development of an immunotherapeutic molecule, a humanized immunotoxin, for treating hCG-expressing advanced-stage cancers. PiPP, high-affinity anti-hCG monoclonal antibody, is used in immunotoxin 'homing' hCG-positive cancer cells. The deimmunized (DI) form α-Sarcin, fungal-origin toxin that lacks functional T-cell epitopes, design to ensure minimal immunogenicity repetitive use humans. A single-chain variable fragment (scFv) PiPP was constructed by linking VH and VL regions antibody. scFv part antibody further linked α-Sarcin at gene level expressed as recombinant protein E. coli. purified from bacterial cell lysate analyzed binding cytotoxicity hCG-secreting colorectal pancreatic results showed scFv(PiPP)-Sarcin able bind cells killed approximately 85% In vivo testing produced similar those vitro against adenocarcinoma-induced tumors.
