Objectives The aim of this study is to characterize the genomic features ST6115 carbapenem-resistant Klebsiella pneumoniae (CRKP) co-harboring bla NDM-1 and IMP-4 . Methods genome sequences one (KP) strain 12 ST17 KP were obtained through whole sequencing (WGS). Subsequently, a phylogenetic analysis was employed ascertain clonal relationship these strains. antimicrobial susceptibility testing (AST) evaluated application minimum inhibitory concentration (MIC) methodology by broth microdilution method. Annotation enabled identification plasmid structure comparative genetic background. Finally, conjugation transfer experiment conducted assess transferability pHD8428-NDM-IMP. Results A comparable revealed that HD8428 majority strains (9/12) clustered on same clade, which suggests close between two ST types. Additionally, showed particularly similarity HD11490, with single-nucleotide polymorphism (SNP) difference only 273. antibiotic resistance genes carried 13 all carry extended-spectrum β-lactamase (ESBL) genes. AST exhibited diverse range antibiotics. identified as being located Further demonstrated gene present ΔTn 125 , while In1377-2. results indicated pHD8428-NDM-IMP may pose risk for transmission in healthcare settings. Conclusion We report clinical identify coexistence IncHI5 type resistant wide antibiotics harbored transmissible pHD8428-NDM-IMP, made it potential threat public health. Our indicates system services should remain vigilant regarding spread prevalence ST6115.

KL1, KL2 CR-hvKP and KL64, KL47 hv-CRKP all exhibit overlapping multidrug resistance hypervirulence phenotypes, but the differences in epidemiological, phenotypic genotypic characteristics between them remains unclear. In this study, we collected non-repeated hv-CRKP/CR-hvKP isolates a tertiary hospital Shanghai, China from January 2019 to December 2022. Furthermore, selected four typical hypervirulent carbapenem-resistant Klebsiella pneumoniae, including ST23-KL1/ST86-KL2 (WYKP3 WYKP194) ST11-KL64/ST11-KL47 (WYKP589 WYKP188), tried clarify compare their virulence drug plasmid distribution. Our study found that ST23-KL1 ST86-KL2 exhibited less diversity than of ST11-KL64 ST11-KL47 hv-CRKP. Compared with hv-CRKP, harbored significantly fewer antimicrobial genes more genes, which contributed higher these strains antibiotics. has emerged as most prevalent pneumoniae probably due its clonal transmission within hospitals well plasmids help conjugative plasmids. Due different evolutionary mechanisms capsular serotypes, are different.

ABSTRACT Carbapenem-resistant Morganella morganii (CRMM) isolates, particularly those producing Klebsiella pneumoniae carbapenemase-2 (KPC-2) or New Delhi metallo-β-lactamase-1 (NDM-1), are increasingly being recognized as causative agents of nosocomial infections. However, systematic phylogeography and genetic characterization these isolates worldwide still lacking. Here, through seven years surveillance CRMM in a tertiary hospital, we analyzed the genomic characteristics bla KPC-2 - NDM-1 -positive isolates. Furthermore, conducted global epidemiological study spp. harboring KPC NDM using NCBI database over past decade. By combining timeline isolate collection with structural analysis plasmids, traced evolution IncL/M plasmid, which acquired gene. Our findings indicate that horizontal transfer Tn 6296 based on IS 26 is crucial for transmission Additionally, 125 transposon appears to have played an important role early plasmid-mediated dissemination ; however, it has been surpassed recent by other elements, including CR . In summary, phylogeographic globally, elucidated their spatial–temporal distribution revealed evolutionary KPC- NDM-producing predominant "epidemic" clone. IMPORTANCE Currently, infections attributable carbapenem-resistant rise, highlighting increasing severity antimicrobial resistance. this study, trajectory genes within core environments. We emphasize necessity strengthening controlling organisms clinical settings prevent generation so-called "superbug"

The emergence of carbapenem-resistant Escherichia coli (CREC) poses crucial challenges in clinical management, requiring continuous monitoring to inform control and treatment strategies. This study aimed investigate the genomic epidemiological characteristics CREC isolates obtained from a tertiary hospital China between 2015 2022. Next-generation sequencing was used for profiling, data patients were integrated into analysis. ST405 (21.2%), ST167 (20.3%) ST410 (15.9%) most prevalent 30 distinct sequence types (STs) identified among 113 unique isolates. Infections caused by clone severe underlying diseases associated with higher in-hospital mortality rates, particularly aged ≥65 years. Furthermore, exhibited greater number virulence resistance genes than non-ST405 clones. gene eaeX mph(E) msr(E) exclusively found clones, while other (agn43, ipad malX) (armA, catB3 arr-3) more this Additionally, showed minimum inhibitory concentrations both meropenem imipenem superior growth under challenge. Galleria mellonella assays revealed that virulent predominant STs. Our findings underscore threat posed clone, which exhibits enhanced extensive drug resistance. These results highlight urgent need stringent surveillance targeted interventions curb its further dissemination prevent potential outbreaks.
