Alzheimer's disease (AD) is one of the major neurological disorders causing death in elderly worldwide. As a neurodegenerative that difficult to prevent and cure, pathogenesis AD complex there no effective cure. A variety natural products derived from plants have been reported promising anti-AD activities, including flavonoids, terpenes, phenolic acids alkaloids, which can effectively relieve symptoms ways. This paper mainly reviews pharmacological activity mechanisms against AD. Although clinical efficacy these still needs be determined by further high-quality studies, it may also provide basis for future researchers study depth.

Abstract To investigate the effect of dipeptidyl peptidase 4 (DPP4) on cognitive impairment in Alzheimer's disease (AD), present study used seven-week-old male C57BL/6J and DPP4 knockout mice. The AD model was induced by microinjection Aβ25−35 into lateral ventricle. Morris water maze test showed that significantly improved spatial learning memory abilitoes Western blot results increased expression levels BDNF, CREB Bcl-2 hippocampus mice while Caspase-8, pyroptosis-related proteins NLRP3, Caspase-1, GSDMD, 1L-18, 1L-1β apoptosis-related Caspase-3 Bax were decreased. Similar observed after HT22 neurons treated with inhibitor sitagliptin (Sit). Moreover, treatment Caspase-8 (Z-LETD-FMK) inhibition inhibited NLRP3 Caspase-1 cells, but had no further inhibitory under Sit. Our suggests may ameliorate dysfunction regulating pyroptosis apoptosis pathways through Caspase-8.

Alzheimer's disease, is one of the irreversible neurodegenerative disorders among older people, causes behavior, memory, and thinking problems. This study aim to synthesize 4‐({8‐[3‐(dimethylamino)phenoxy]octyl}oxy)phthalonitrile, zinc(II) phthalocyanine its water soluble derivative containing ({8‐[3‐(dimethylamino)phenoxy]octyl}oxy) substituents at peripheral positions first time. To determine potential silicon(IV) phthalocyanines for use in AD, spectrophotometric assays were performed their acetylcholinesterase/butyrylcholinesterase inhibitory effects. In addition, DNA nuclease properties evaluated by agarose gel electrophoresis. The results showed that compounds inhibited acetylcholinesterase/butyrylcholinesterase. They did not damage pBR322 plasmid depending on time concentration. As a result experiments, it revealed may be as cholinesterase inhibitors, which promising approach treatment disease.

: Alzheimer’s disease is a multifaceted neurodegenerative disease. Cholinergic dysfunction, amyloid β toxicity, tauopathies, oxidative stress, neuroinflammation are among the main pathologies of Ligands targeting more than one pathology, multi-target directed ligands, attract attention in recent years to tackle In this review, we aimed cover different biochemical pathways, that revealed for pathology disease, as druggable targets such cannabinoid receptors, matrix metalloproteinases, histone deacetylase and various kinases including, glycogen synthase kinase-3, mitogen-activated protein kinase c-Jun N-terminal kinase, their ligands treatment hope providing realistic insights into field.
