Abstract Background Rising global mortality due to antibiotic-resistant pathogens necessitates novel antibacterial and antifungal agents. This study focuses on synthesizing gold nanoparticles (GNPs) via tricyclic microwave irradiation (TMI) combat Multi-Drug-Resistant Bacteria Fungus. The demand for sustainable synthesis methods has led the exploration of TMI GNP production. Results Characterization demonstrates consistent, uniform, dispersed GNPs with trigonal hexagonal shapes. sized 20–55 nm exhibit superior activity, particularly against drug-resistant Gram-positive bacteria. Notably, display consistent efficacy fungus demonstrate potential broad-spectrum antimicrobial applications. Conclusion TMI-synthesized GNPs, characterized by their favorable physical properties size-dependent efficacy, show promise as effective agents pathogens. Rhino-orbito-cerebral mucormycosis (ROCM) is a rare and life-threatening fungal infection that predominantly affects immunocompromised individuals, such as those undergoing hematopoietic stem cell transplantation (HSCT). This report describes the case of 10-year-old girl with very severe aplastic anemia (VSAA) who underwent an haploidentical HSCT from her father. She initially achieved successful engraftment donor chimerism rate 98.25% on day 60. However, 65 post-transplant, she developed right-eye pain, progressive swelling, visual impairment. Comprehensive evaluations, including contrast-enhanced magnetic resonance imaging (MRI) cerebrospinal fluid (CSF) analysis, revealed extensive orbital cerebral involvement consistent ROCM. Mucormycosis is a highly invasive and rare opportunistic infection caused by mucor fungi, characterized challenging diagnosis rapid disease progression. Dual-target (CYP51/PD-L1) plays an important role in the process of fungal proliferation and immune suppression. A series novel quinazoline compounds with dual-target inhibition function was constructed using skeleton growth method, their structures were synthesized, characterized, evaluated. Among them, perfected (L11, L20, L21) selected for further study, which exhibited remarkable biological activity against different strains (MIC50, 0.25-2.0 μg/mL) vitro. On one hand, these inhibited CYP51 activity, induced ROS aggregation, mitochondrial damage; this ultimately caused lysis death. other they also effectively activated body's ability by blocking interaction between PD-L1 PD-1, slowed down inflammatory reaction, accelerated recovery infections.

In this study, PD-L1 and CYP51 were selected as key dual-target enzymes, which play an important role in the process of fungal proliferation immune suppression. Faced with increasingly serious fungal infections and drug resistance issues, three different series of novel dual-target (programmed death ligand 1/14 α-demethylase) compounds were constructed through the fragment combination pathway in study. Their chemical structures synthesized, characterized, evaluated. Among them, preferred 10c-1, 17b-1, 18b-2 could efficiently exert their antifungal antidrug-resistant ability blocking ergosterol biosynthesis, inducing upregulation reactive oxygen species level, triggering apoptosis. Especially, compound exhibited synergistic function inhibition immune activation. Moreover, covalent organic framework carrier was also generated based on acidic microenvironment infection to improve bioavailability targeting compounds; this finally accelerated body's recovery rate.

The rise in cases (COVID-19-associated mucormycosis, CAM) is attributed to excessive steroid and antibiotic use, poor hospital hygiene, crowded settings. Major contributing factors include diabetes weakened immune systems. main manifesting forms of CAM─cutaneous, pulmonary, deadliest, rhinocerebral─and disseminated infections elevated mortality rates 85%.
