Osteoarthritis (OA) is the most common degenerative joint disease. Multiple tissues are altered during development of OA, resulting in pain and permanent damage to osteoarticular joints. Current research has demonstrated that non-apoptotic cell death plays a crucial role OA. With continuous targeted therapies, shown great potential prevention treatment We systematically reviewed progress on pathogenesis, development, outcome including autophagy, pyroptosis, ferroptosis, necroptosis, immunogenic death, parthanatos. This article reviews mechanism OA provides theoretical basis for identification new targets treatment.

Necroptosis is a regulated inflammatory cell death process that closely associated with autoimmune diseases, acute ischemic injuries, neurodegenerative disorders, and so on. Due to the crucial role of receptor-interacting protein kinase 1 (RIPK1) in necroptosis pathway, RIPK1 inhibitors are believed have great potential treatment necroptosis-related diseases. In this article, we reported series pyridazin-4-one derivatives as potent for both human mouse cells. The representative compound 13 exhibited favorable selectivity dose-dependently inhibited phosphorylation. vivo pharmacokinetic study indicated was an orally available candidate. Finally, molecular docking dynamics simulations were performed elucidate binding pattern RIPK1. Collectively, represents promising lead future development RIPK1-targeted inhibitors.
