Alzheimer’s disease (AD), is a progressive neurodegenerative that affects behavior, thinking, learning, and memory in elderly individuals. AD occurs two forms, early onset familial late-onset sporadic; genetic mutations PS1, PS2, APP genes cause AD, combination of lifestyle, environment factors causes the sporadic form disease. However, accelerated progression noticed patients with AD. Disease-causing pathological changes are synaptic damage, mitochondrial structural functional changes, addition to increased production accumulation phosphorylated tau (p-tau), amyloid beta (Aβ) affected brain regions patients. Aβ peptide derived from precursor protein (APP) by proteolytic cleavage gamma secretases. glycoprotein plays significant role maintaining neuronal homeostasis like signaling, development, intracellular transport. reported have both protective toxic effects neurons. The purpose our article summarize recent developments its association synapses, mitochondria, microglia, astrocytes, interaction p-tau. Our also covers therapeutic strategies reduce toxicities discusses reasons for failures therapeutics.

Flavonoids are among the most abundant natural bioactive compounds produced by plants. Many different activities have been reported for these secondary metabolites against numerous cells and systems. One of interesting is certainly antimicrobial, which stimulated through various molecular mechanisms. In fact, flavonoids effective both in directly damaging envelope Gram-negative Gram-positive bacteria but also acting toward specific targets essential survival microorganisms. The purpose this paper to present an overview results obtained research focused on study interactions between bacterial proteins. Despite great structural heterogeneity plant metabolites, it observe that many affect same cellular pathways. Furthermore, evident some interact with more than one target, producing multiple effects. Taken together, data demonstrate potential developing innovative systems, can help address increasingly serious problem antibiotic resistance.

Alzheimer disease (AD) is associated with multiple etiologies and pathological mechanisms, among which oxidative stress (OS) appears as a major determinant. Intriguingly, OS arises in various pathways regulating brain functions, it seems to link different hypotheses mechanisms of AD neuropathology high fidelity. The particularly vulnerable damage, mainly because its unique lipid composition, resulting an amplified cascade redox reactions that target several cellular components/functions ultimately leading neurodegeneration. present review highlights the “OS hypothesis AD,” including amyloid beta-peptide-associated role protein oxidation unraveled by proteomics, antioxidant strategies have been investigated modulate progression AD. Collected studies from our groups others contributed unraveling close relationships between perturbation homeostasis elucidating redox-regulated events potentially involved both pathogenesis However, complexity requires in-depth understanding intracellular affecting relevant for functions. This crucial developing pharmacological targeting OS-mediated toxicity may contribute slow well improve quality life persons this severe dementing disorder.

Neurodegenerative diseases, such as Alzheimer's disease (AD), are characterized by massive loss of specific neurons. It is a progressive disabling, severe and fatal complex disease. Due to its pathogenesis limitations clinical treatment strategies, it poses serious medical challenge burden worldwide. The AD not clear, potential biological mechanisms include aggregation soluble amyloid form insoluble plaques, abnormal phosphorylation tau protein formation intracellular neurofibrillary tangles (NFT), neuroinflammation, ferroptosis, oxidative stress metal ion disorders. Among them, ferroptosis newly discovered programmed cell death induced iron-dependent lipid peroxidation reactive oxygen species. Recent studies have shown that closely related AD, but the mechanism remains unclear. may be iron metabolism, amino acid metabolism affecting accumulation ions. Some chelating agents (deferoxamine, deferiprone), chloroiodohydroxyquine derivatives, antioxidants (vitamin E, lipoic acid, selenium), derivatives Fer-1, tet, etc. been in animal effective exert neuroprotective effects. This review summarizes regulation natural plant products on order provide reference information for future research development inhibitors.

Alzheimer’s disease (AD) is a progressive neurodegenerative disorder that eventually leads to dementia and death of the patient. Despite enormous amounts resources efforts for AD drug development during last three decades, no effective treatments have been developed can slow or halt progression disease. Currently available drugs treating only improve clinical symptoms temporarily with moderate efficacies. In recent years, scientific community has realized these challenges reconsidered future directions development. The most significant changes in strategy include shifting from amyloid-based targets other targets, such as tau, toward better designs trials. However, still focused on single mechanism target, which conventional Although multifactorial mechanisms and, this basis, multi-target strategies proposed approach not widely recognized accepted by mainstream Here, we emphasize discuss urgent need paradigm shift target multiple either multi-target–directed ligands combination therapy approach. In this research, various green extraction techniques were employed to recover molecules from Kalanchoe daigremontiana (kalanchoe), epicarp Cyphomandra betacea (tamarillo), cooperage woods Robinia pseudoacacia (acacia) Nothofagus pumilio (lenga), as well reference extract (positive control) Rosmarinus officinalis L . (rosemary). The capacity these plant extracts was evaluated in set vitro assays, including enzymatic [acetylcholinesterase (AChE), butyrylcholinesterase (BChE), lipoxygenase (LOX)] antioxidant [ABTS, reactive oxygen nitrogen species (ROS RNS)] bioactivity tests. Extracts also submitted parallel artificial membrane permeability assay mimicking the blood–brain barrier (PAMPA-BBB) two cell viability assays HK-2 SH-SY5Y lines. Comprehensive phytochemical profiling based on liquid chromatography coupled quadrupole-time-of-flight mass spectrometry (LC-Q-TOF-MS) analysis showed enriched content phenolic terpenoid target extracts. Moreover, tests promising capacity, particularly for supercritical-fluid (SFE) acacia (ABTS IC 50 = 0.11 μg ml −1 ; ROS 1.56 AChE 4.23 BChE 1.20 LOX 4.37 ), whereas PAMPA-BBB revealed high perfusion some representative compounds, such acids or flavonoids. Regarding cytotoxic tamarillo rosemary SFE can be considered non-toxic, lenga pressurized (PLE) mild-cytotoxic, kalanchoe highly toxic obtained results demonstrate great potential studied biomass transformed into valuable food additives, supplements, nutraceuticals
