Colorectal cancer (CRC) remains one of the leading causes cancer-associated mortality worldwide. While immune checkpoint inhibitors have shown promise in treatment, there is a need for reliable biomarkers to predict patient prognosis and guide personalized therapies. Palmitoylation, post-translational modification, has been implicated various processes, yet its role CRC unclear. Transcriptome, survival, somatic mutation copy number variation data were retrieved from The Cancer Genome Atlas, GSE17538 dataset was used external validation. A palmitoylation-related risk signature developed using univariate Cox regression, Least Absolute Shrinkage Selection Operator multivariate regression analyses. Patients stratified into high- low-risk groups based on median score. Prognostic accuracy assessed receiver operating characteristic curves Kaplan-Meier overall survival (OS) analysis with validation an independent cohort. Functional enrichment, cell infiltration drug sensitivity analyses performed explore underlying mechanisms therapeutic implications. Subsequently, keratin 8 pseudogene 12 (KRT8P12) overexpression evaluated HT29 cells, knockdown lines generated lentivirus. Cell proliferation Counting Kit-8 5-ethynyl-2'-deoxyuridine assays, migration by Transwell assay apoptosis Annexin-V/propidium iodide staining. consisting six genes (KRT8P12, ZDHHC3, PCOLCE2, MPP2, LARS2 MMAA) identified. High-risk patients exhibited significantly worse OS (HR=3.19; P<0.001) compared patients. Immune revealed enhanced activity group, which associated higher expression genes. Immunotherapy prediction models indicated that might benefit more inhibitors. Drug identified distinct response profiles between groups. Furthermore, vitro, KRT8P12 promoted tumor while inhibiting apoptosis. In conclusion, present study confirmed as gene regulating CRC. addition, palmitoylation-associated may offer promising tool prognostic could treatment strategies, including targeted

Purpose: The purpose of this research is to explore how Orange, a powerful information extraction and predictive modeling software, can be applied in the pharmaceutical industry assess visualize effectiveness cancer prevention medicines. By focusing on companies like Genentech Inc. (USA), AstraZeneca Pharmaceutical PLC (UK), Boehringer Ingelheim (Germany), Chugai Co. Ltd. (Japan), study seeks evaluate which cancer-preventing drugs from these provide best efficacy while minimizing side effects for patients. goal assist healthcare professionals (doctors pharmacists) making informed decisions about most suitable medications prevention, ensuring patient safety optimal treatment outcomes. Design/Methodology/Approach: This utilizes Orange software’s machine learning data visualization tools, specifically scatterplot graphs, analyze complex datasets related drugs. using scatterplots concurrently examine multiple parameters, such as Company Name, Drug Class, Medicine Prevention Cancer Diseases, Side Effects Percentage aims identify patterns correlations that help drug safety. approach involves analyzing relationship between characteristics effects, providing actionable insights into different treatments might interact with health conditions. Findings/Results: findings suggest Orange’s visualizations valuable various medicines across companies. enabling simultaneous analysis software helps are effective preventing effects. provides clearer understanding characteristics, outcomes, supporting data-driven decision-making development practices. Originality/Value: originality lies application mining capabilities relationships within datasets. use efficacy, an innovative offers richer, more nuanced contributes optimizing choice strategies, ultimately improving therapeutic Paper Type: analytical paper applies techniques focuses tools extract interpret data, professionals.
