With numerous novel and innovative in vitro models emerging every year to reduce or replace animal testing, there is an urgent need align the design, harmonization, validation of such systems using vitro-in vivo extrapolation (IVIVE) approaches. In particular, inhalation toxicology, a lack predictive prevalidated lung that can be considered valid alternative for testing. The power enhanced by applying Adverse Outcome Pathways (AOP) framework, which casually links key events (KE) relevant IVIVE. However, one difficulties identified endpoint analysis readouts specific assays toxicants are currently not harmonized, making alignment challenging. We summarize current state art two systems, focusing on inflammatory-induced effects providing guidance future research directions improve alignment.
