Microplastic (MP) pollution has become a global environmental issue, and increasing concern been raised about its impact on human health. Current studies the toxic effects mechanisms of MPs have mostly conducted in animal models or vitro cell cultures, which limitations regarding inter-species differences stimulation cellular functions. Organoid technology derived from pluripotent adult stem cells broader prospects for predicting potential health risks to humans. Exposure to micro- and nanoplastics (MNPs) has been implicated in potential cardiotoxicity. However, vitro models based on cardiomyocyte cell lines lack crucial cardiac characteristics, while interspecies differences animal compromise the reliability of conclusions. In relation to the eye, body can absorb substances from ocular surface fluid (OSF) in a few ways: directly through conjunctival sac, nasal mucosa as drains into nose, or ingestion. Regardless of absorption method, sac should be used toxicological matrix, even though only small quantities are needed. Contemporary analytical techniques make it suitable matrix for research. Organ-chip technology, in contrast to cell culture and animal models, offers a promising platform for accelerating drug development. However, current chip designs simulate human organ functions there is lack of multi-organ that can simultaneously study efficacy hepatorenal toxicity. Here, we developed novel microfluidic integrated cholangiocarcinoma organoids (CCOs) with recellularized liver slices (RLS) kidney (RKS), assess anti-cholangiocarcinoma Co-culture patient-derived CCOs RLS RKS was successfully achieved 7 days under flow conditions enhanced renal functions. Furthermore, an vitro biomimetic model showed IC50 values trastuzumab emtansine (T-DM1) around 6.42 ± 7.34 μg mL-1 four clinical cases, one outlier 77.77 due patient variability. Post-treatment, viability remained high at 75.67% 81.03%, respectively, suggesting low toxicity T-DM1 treating cholangiocarcinoma. Our demonstrates the use organoid-slice-on-a-chip (OSOC) personalized assessment, particularly aiming leveraging anticancer drugs off-label save lives.
