Bisphenol A, an endocrine-disrupting compound, is widely used in the industrial production of plastic products. Despite increasing concerns about its harmful effects on human health, animals, and environment, use BPA has been banned only infant products, cellular processes are not fully understood. To investigate impact eukaryotic cells, we analyzed proteome changes wild-type PDR5-deleted S. cerevisiae strains exposed to different doses using sample multiplexing-based proteomics. We found that ABC multidrug transporter Pdr5 plays important role protecting yeast cells from toxicity, with absence significantly sensitizing BPA. inhibited growth a dose-dependent manner, more pronounced effect cells. Proteomic analysis revealed induces widespread protein abundance, including upregulation metabolic pathways such as arginine biosynthesis downregulation mitochondrial proteins. Additionally, observed markers stress induced by identifying multiple stress-induced proteins were upregulated this compound. As affected have shown be evolutionarily conserved, these insights can advance our understanding BPA's broader health.

Bisphenol analogues have been shown to similar estrogenic activity that of BPA and may affect fetal development. However, no human studies examined the effects perinatal exposure emerging bisphenol alternatives [bisphenol G, M, BP (BPBP)] on small for gestational age (SGA) how placental function mediate relationship. Here, 13 urinary were detected in 1054 contemporary pregnant women, was still most dominant congener. Logistic regressions identified its traditional B (BPB), E (BPE), Z, AP (BPAP)] as being associated with an elevated risk SGA (all ORs > 1.80, P < 0.05). In contrast, substitutes, despite high occurrences, all showed much attenuated risk. Mixture effect models Bayesian kernel machine regression quantile-based g-computation demonstrated coexposure bisphenols strongly correlated (OR = 2.70, 0.001), conventional substitutes (BPB, BPE, BPAP) primary drivers, outweighing from substitutes. Finally, mediation analysis revealed index estriol mediated relationship between SGA, dominated by BPBP (25.4%). Our findings provide new epidemiological evidence early pose a higher offspring development than those alternatives, potentially via compromised function. Future toxicity assessments validation other settings these are needed.

Bisphenol A (BPA), a widely used chemical in the production of plastics and epoxy resins, has garnered significant attention due to its association with adverse health effects, particularly endocrine-disrupting properties. Regulatory measures aimed at reducing human exposure BPA have led proliferation alternative chemicals various consumer industrial products. While these alternatives serve reduce exposure, concerns arisen regarding their safety potential toxicity as regrettable substitutes. Previous efforts demonstrated that vitro high-throughput transcriptomics (HTTr) studies can be assess alternatives, this strategy produces transcriptomic points-of-departure (tPODs) are protective when compared PODs from traditional rodent studies. In study, we HTTr for eleven data-poor legacy sharing structural similarities BPA. Human breast cancer MCF-7 cells were exposed 11 concentrations ranging 0.1 25 μM toxicity. Analysis global changes previously characterized estrogen receptor alpha (ERα) biomarker signature revealed 9 altered gene expression relative controls. One (2,4'-Bisphenol A) activated ERα same concentration (i.e., 4,4'-BPA) but was deemed more potent it induced lower concentrations. These results address data gaps support ongoing screening assessments identify hazard help candidates may safer alternatives.
