In the present research work, state-of-art label-free electrochemical genosensing platform was developed based on hybridization process in presence of [Fe(CN)6]3−/4− as an efficient redox probe for sensitive recognition miRNA-21 human gastric cell lines samples. To attain this aim, perovskite nanosheets were initially synthesized. Afterward, obtained compound combined with graphene oxide resulting effective modifier, which dropped surface Au electrode. Then, AuNPs (Gold Nano Particles) have been electrochemically-immobilized perovskite-graphene oxide/Au-modified electrode through chronoamperometry (CA) technique. Finally, a self-assembling monolayer reaction ss-capture RNA ensued by thiol group at end modified surface. has cast to apply process. find out effectiveness synthesized modifier agent, behavior analyzed DPV (differential pulse voltammetry) and CV (cyclic techniques. The prepared biomarker-detection bioassay offers high sensitivity specificity, good performance, appropriate precision accuracy highly-sensitive determination miRNA-21. Different characterization methods used, such XRD, Raman, EDS, FE-SEM, morphological investigation particle size. Based optimal conditions, limit detection quantification acquired 2.94 fM 8.75 fM, respectively. Furthermore, it possible achieve wide linear range is between 10−14 10−7 Moreover, selectivity proposed biosensing assay investigated its potential one, two, three-base mismatched sequences. investigate repeatability reproducibility related bio-assay. evaluate process, important that planned biomarker bio-assay could be directly re-used re-generated.
