Dry eye disease (DED) is a multifactorial condition characterized by ocular surface inflammation, tear film instability, and corneal epithelial damage. Current treatments often provide temporary relief without addressing the underlying inflammatory mechanisms. This study examined therapeutic potential of crocin nobiletin, two naturally derived compounds with well-known antioxidant anti-inflammatory properties, in mouse model DED induced lacrimal gland excision (LGE). Thirty female Balb/c mice were divided into five groups (n = 6 each): Control (sham surgery), untreated DED, nobiletin-treated (32.75 µM), crocin-treated (34 1% betamethasone-treated DED. Treatments administered three times daily for 28 days. Ocular tissues evaluated using Hematoxylin Eosin (H&E) staining fluorescein staining. Conjunctival cytokines, including interleukin-6 (IL-6), interleukin-1 beta (IL-1β), tumor necrosis factor-alpha (TNF-α), measured enzyme-linked immunosorbent assay (ELISA). Histological analysis showed that nobiletin treatment exhibited reduced disruption, keratinization, cell infiltration compared to group. The ELISA revealed both efficiently inhibited production pro-inflammatory cytokines IL-6, TNF-α, IL-1β, which are key mediators pathogenesis. Fluorescein further confirmed protective impact on integrity. Moreover, epithelial-preserving effects these comparable those corticosteroid betamethasone. Dry eye syndrome (DES) is a chronic ocular disease that induces epithelial damage to the cornea by decreasing tear production and quality. Adequate treatment options have not been established for severe DES such as Sjogren's due complicated pathological conditions. To solve this problem, we focused on conditioned medium of human adipose-derived mesenchymal stem cells (hAdMSC-CM), which multiple therapeutic properties. Here, showed hAdMSC-CM suppressed Benzalkonium Chloride (BAC)-induced cytotoxicity inflammation in corneal (hCECs). In addition, increased expression level regulated localisation barrier function-related components, improved BAC-induced dysfunction hCECs. RNA-seq analysis pharmacological inhibition experiments revealed effects were associated with TGFβ JAK-STAT signalling pathways. Moreover, model rats exorbital intraorbital lacrimal gland excision, instillation improving cornea. Thus, demonstrated has properties pathways, may serve an innovative agent function.

Abstract The insights into how genome topology couples with epigenetic states to govern the function and identity of corneal epithelium are poorly understood. Here, we generate a high-resolution Hi-C interaction map human limbal stem/progenitor cells (LSCs) show that chromatin multi-hierarchical organisation is coupled gene expression. By integrating Hi-C, epigenome transcriptome data, characterize comprehensive 3D epigenomic landscapes LSCs. We find super-silencers mediate repression associated development, differentiation disease via looping and/or proximity. Super-enhancer (SE) analysis identified set SE interactive hubs contribute LSC-specific activation.
