This study aimed to identify the serum metabolites associated with sarcopenic risk in Japanese patients type 2 diabetes, determine effect of dietary protein intake on metabolic profile, and examine its association sarcopenia. Ninety-nine diabetes were included, was defined as low muscle mass or strength. Seventeen quantified after gas chromatography–mass spectrometry analysis. Background Untargeted metabonomics has provided new insight into the pathogenesis of sarcopenia. In this study, we explored plasma metabolic signatures linked to a heightened risk sarcopenia in cohort study by LC-MS-based untargeted metabonomics. Methods nested case–control from Adult Physical Fitness and Health Cohort Study (APFHCS), collected blood samples 30 new-onset subjects (mean age 73.2 ± 5.6 years) healthy controls 74.2 4.6 matched age, sex, BMI, lifestyle, comorbidities. ABSTRACT Sarcopenia (SP), an age‐associated condition marked by muscle weakness and loss has been strongly connected with metabolic factors according to substantial evidence. Nevertheless, the causal correlation between SP serum metabolites, biological signaling pathways involved, is still not well understood. We performed a bidirectional two‐sample Mendelian randomization (MR) analysis examine relationships 1091 levels 309 ratios of metabolites traits, alongside investigating relevant pathways. The intricate relationship between bile acid metabolism and skeletal muscle function has emerged as a crucial area of research in metabolic health. This review synthesizes current evidence highlighting the fundamental role acids key signaling molecules homeostasis their therapeutic potential muscle-related disorders. Recent advances molecular biology metabolomics have revealed that acids, beyond classical lipid absorption, essential regulators mass through multiple pathways, particularly via nuclear receptor FXR membrane TGR5. Clinical studies demonstrated significant associations altered profiles wasting conditions, while experimental elucidated underlying mechanisms linking to protein synthesis, energy metabolism, regeneration capacity. Background: Walking ability is important for the quality of life older adults. A self-reported walking index (WAI) covering difficulty and ease captures a broader spectrum in healthy persons. Methods: Using metabolomics Health, Aging Body Composition study, we identified Year 2 metabolites cross-sectionally longitudinally related to WAI (0–9, higher scores indicate better ability) using probabilistic models multinomial logistic models, respectively. Results: Among 2334 participants (mean age 74.6 years, 51% women, 37% Black), 27% scored 0–5, 36% 6–8, 9 at 2. Over 4 52% maintained stable WAI, 6% improved, while 42% declined (22% 1–2 points 20% >2 decline). We 81 significantly associated with both poorer concurrent faster decline, including acylcarnitine species, shorter-chain saturated diglycerides triglycerides, TCA cycle intermediates (cis-aconitic, fumaric, malic acids), lower phospholipids levels.
