Functional Analysis of Durum Wheat GASA1 Protein as a Biotechnological Alternative Against Plant Fungal Pathogens and a Positive Regulator of Biotic Stress Defense

Plants
are
frequently
challenged
by
a
variety
of
microorganisms.
To
protect
themselves
against
harmful
invaders,
they
have
evolved
highly
effective
defense
mechanisms,
including
the
synthesis
numerous
types
antimicrobial
peptides
(AMPs).
Snakins
such
compounds,
encoded
GASA
(Gibberellic
Acid-Stimulated
Arabidopsis)
gene
family,
and
involved
in
response
to
biotic
abiotic
stress.
Here,
we
examined
function
newly
identified
TdGASA1
its
protein
Triticum
durum
subjected
different
stress-related
simulants,
as
mechanical
injury,
methyl
jasmonate
(MeJA),
indole-3-acetic
acid
(IAA),
salicylic
(SA),
hydrogen
peroxide
(H2O2),
well
infection
with
pathogenic
fungi
Fusarium
graminearum
Aspergillus
niger.
We
found
that
wheat,
transcripts
were
markedly
increased
these
stress
simulants.
Isolated
purified
exhibited
significant
antifungal
activity
growth
inhibition
test
conducted
on
eight
species
solid
liquid
media.
Transgenic
Arabidopsis
lines
overexpressing
obtained
this
study
showed
higher
tolerance
detrimental
effects
H2O2,
MeJA,
ABA
treatment.
In
addition,
resistance
niger,
which
was
linked
marked
increase
antioxidant
leaves
under
conditions.
This
correlated
upregulation
pathogenesis-related
genes
(AtPDF1.2a,
AtERF1,
AtVSP2,
AtMYC2,
AtPR1,
AtACS6,
AtETR1,
AtLOX2)
transgenic
lines.
Overall,
our
results
indicate
respond
ubiquitously

Essential
oils
(EOs)
from
various
medicinal
and
aromatic
plants
are
known
for
their
diverse
biological
activities,
including
antimicrobial
effects.
Citrus
aurantium
EO
is
traditionally
used
therapeutic
benefits
due
to
its
high
content
of
bioactive
compounds.
Therefore,
this
study
focuses
on
potential
use
as
a
food
preservative
by
investigating
the
combined
antibacterial
properties
EOs
leaves
(EO1),
flowers
(EO2),
small
branches
(EO3)
against
six
bacterial
strains
agar
disk
diffusion,
minimum
inhibitory
concentration
(MIC),
bactericidal
(MBC)
methods.
The
chemical
compositions
were
analysed
gas
chromatography–mass
spectrometry
(GC-MS)
revealed
presence
numerous
compounds
responsible
properties.
