Cardiovascular diseases (CVDs) caused by arteriosclerosis are the leading cause of death and disability worldwide. In late stages atherosclerosis, atherosclerotic plaque gradually expands in blood vessels, resulting vascular stenosis. When unstable ruptures falls off, it blocks vessel causing thrombosis, to strokes, myocardial infarctions, a series other serious that endanger people's lives. Therefore, regulating stability is main means used address high mortality associated with CVDs. The progression complex integration cell apoptosis, lipid metabolism disorders, inflammatory infiltration, smooth muscle migration, neovascular infiltration. More recently, emerging evidence has demonstrated non-coding RNAs (ncRNAs) play significant role pathophysiological process formation affecting biological functions vasculature its cells. purpose this paper comprehensively review regulatory mechanisms involved susceptibility rupture, discuss limitations current approaches treat instability, highlight potential clinical value ncRNAs as novel diagnostic biomarkers therapeutic strategies improve reduce risk major cardiovascular events.

Abstract Knowledge regarding the relationship between molecular mechanisms underlying atherosclerosis (AS) and transfer RNA‐derived small RNAs (tsRNAs) is limited. This study illustrated expression profile of tsRNAs, thus exploring its roles in AS pathogenesis. Small RNA sequencing was performed with four atherosclerotic arterial healthy subject samples. Using bioinformatics, protein‐protein interaction network cellular experiments were constructed to predict enriched signalling pathways regulatory tsRNAs AS. Of total 315 identified be dysregulated group, 131 184 up‐regulated down‐regulated, respectively. Interestingly, pathway differentiated cell adhesion molecules (CAMs) implicated closely associated Particularly, tRF‐Gly‐GCC might participate pathogenesis via regulating adhesion, proliferation, migration phenotypic transformation HUVECs VSMCs. In conclusion, help understand better. may a promising target for suppressing abnormal vessels functions, suggesting novel strategy preventing progression atherosclerosis.

Aminoacyl-tRNA synthetases (ARSs) are widely found in organisms, which can activate amino acids and make them bind to tRNA through ester bond form the corresponding aminoyl-tRNA. The classic function of ARS is provide raw materials for protein biosynthesis. Recently, emerging evidence demonstrates that ARSs play critical roles controlling inflammation, immune responses, tumorigenesis as well other important physiological pathological processes. With recent development genome exon sequencing technology, discovery new clinical cases, have been reported be closely associated with a variety cardiovascular diseases (CVDs), particularly angiogenesis cardiomyopathy. Intriguingly, aminoacylation was newly identified modify substrate proteins, thereby regulating activity functions. Sensing availability intracellular related regulation cell physiology. In this review, we summarize research progress on mechanism CVDs caused by abnormal introduce phenotypes characteristics dysfunction. We also highlight potential CVDs. Finally, discuss some limitations challenges present research. current findings suggest significant involved CVDs, values novel diagnostic therapeutic targets CVD treatment.

Long noncoding RNAs (lncRNAs) are important regulators of various cellular functions. Recent studies have shown that a novel lncRNA termed Punisher is highly expressed in cardiovascular progenitors and has potential role diseases. However, its role, especially molecular mechanism, unclear. In our present study, we observed was obviously downregulated atherosclerotic plaques. Further research proved it can suppress the apoptosis VSMCs potentially contributing to progression atherosclerosis. Intriguingly, revealed regulate mitochondria fission as well mitochondrial functions induced by hydrogen peroxide (H2O2) VSMCs. Mechanistically, further serve ceRNA which directly binds miR-664a-5p consequently regulates target OPA1, finally contributes biological function Particularly, overexpression distinctly suppressed neointima formation VSMC vivo. Encouragingly, these results were accordance with findings obtained clinical evaluation patients Our data provides significant relationship among homeostasis, apoptosis, And could targets diagnosis treatment

Abstract Atherosclerosis (AS), a chronic sterile inflammatory disorder, is one of the leading causes mortality worldwide. The dysfunction and unnatural death plaque cells, including vascular endothelial cells (VEC), macrophages, smooth muscle (VSMC), are crucial factors in progression AS. Pyroptosis was described as form cell at least two decades ago. It featured by plasma membrane swelling rupture, lysis, consequent robust release cytosolic contents pro-inflammatory mediators, interleukin-1β (IL-1β), IL-18, high mobility group box 1 (HMGB1). commonly observed initiation development AS, levels pyroptosis-related proteins positively correlated with instability, indicating contribution pyroptosis to atherogenesis. Furthermore, studies have also identified some candidate anti-atherogenic agents targeting pyroptosis. Herein, we summarize research progress understating (1) discovery definition pyroptosis; (2) characterization molecular mechanisms (3) regulatory VEC, macrophage, VSMC, well their potential role AS progression, aimed providing therapeutic targets for prevention treatment

In recent years, the cure rate of acute pancreatitis (AP) has increased gradually, but morbidity recurrent (RAP) not decreased. Patients with RAP have a poor quality life and are more likely to develop into chronic pancreatitis. To investigate risk factors after first attack is very necessary.Patients episode AP admitted Shanghai General Hospital, Jiao Tong University School Medicine from January 2018 May 2021 were included in this retrospective study, follow-up was 3-45 months discharge. The patients information collected medical records including laboratory tests auxiliary inspection their hospitalization. Univariate multivariate Cox regression analysis used explore RAP. Cumulative plotted using Kaplan-Meier curves.A total 592 enrolled study 81 (13.7%) developed Among those patients, majority (67.9%) men, median age 43 years. most common etiology hypertriglyceridemia (38.3%). Multivariate showed that smoking history (p < 0.001), infected pancreatic necrosis occurred during admission = 0.005), high low-density lipoprotein cholesterol (LDLc) level 0.001) significant independent for above had 3-year cumulative recurrence (32.2%, 45.5%, 28.9%, respectively).Smoking history, necrosis, LDLc decisive Attention should be paid factors.

Nicotine contributes to the causation of atherosclerosis, which prominent cellular components are macrophages. Long non-coding RNAs (lncRNAs) play an important role in regulating cell functions such as proliferation, differentiation and programmed death. However, function mechanism lncRNAs nicotine-induced macrophage pyroptosis has not been reported. We screened deferentially expressed human carotid artery plaque (GSE97210) verified them Results showed only LINC01272 was up-regulated a dose-dependent manner The immunofluorescence staining result confirmed that interfering inhibited pyroptosis. Through bioinformatics analysis, dual luciferase reporter gene assay qPCR, we identified miR-515 significantly negatively correlated with expression LINC01272, KLF6 is target miR-515. Furthermore, our results demonstrated LINC01272/miR-515/KLF6 axis meditated In addition, peripheral blood mononuclear cells smoking populations, GSDMD-N, NLRP3, increased, while level reduced. This study nicotine increases competitively bind macrophages, reducing inhibitory effect on its KLF6, ultimately induces
