Abstract Herein, we report a calcium‐catalyzed, one‐pot, four‐component reaction to synthesize diversely substituted pyrroles. This atom‐economy involves sequence of reactions that proceed via in situ formation α ‐ imino ketones, Friedel‐Crafts arylation, aza‐cyclization, and aromatization the single pot. The showed broad substrate scope with good yields. We demonstrated gram‐scale synthesis post‐synthetic modifications.

Discovery of Novel Chiral Indole Derivatives Containing the Oxazoline Moiety as Potential Antiviral Agents for Plants DOI

Potato virus Y (PVY) is an important plant that has spread worldwide, causing significant economic losses. To search for novel structures as potent antiviral agents, a series of chiral indole derivatives containing oxazoline moieties were designed and synthesized their anti-PVY activities evaluated. Biological activity tests demonstrated many compounds exhibited promising absolute configurations obvious distinctions in bioactivities. Notably, compound (S)-4v displayed excellent curative protective efficacy against PVY, with EC50 values 328.6 256.1 μg/mL, respectively, which superior to those commercial virucide ningnanmycin (NNM, 437.4 397.4 respectively). The preliminary mechanism was investigated determine the difference between two enantiomers 4v compounds. Molecular docking indicated stronger binding affinity coating proteins PVY (PVY-CP) (−6.5 kcal/mol) compared (R)-4v (−6.2 kcal/mol). Additionally, can increase chlorophyll content defense-related enzyme more effectively than its enantiomer. Therefore, this study provides basis development agricultural chemicals.

Abstract BACKGROUND Potato virus Y (PVY) is a prominent representative of plant viruses. It can inflict severe damage upon Solanaceae plants, leading to global dissemination and substantial economic losses. To discover new antiviral agents, class planar chiral thiourea molecules through the key step N ‐heterocyclic carbene‐catalyzed nitrile formation reaction was synthesized with excellent optical purities for evaluations against PVY. RESULTS The absolute configurations compounds exhibited obvious distinctions in anti‐PVY activities. Notability, compound ( S )‐ 4u remarkable curative activities PVY, half maximal effective concentration (EC 50 ) 349.3 μg mL −1 , which lower than that ningnanmycin (NNM) = 400.8 ). Additionally, EC value protective effects 146.2 superior NNM (276.4 Furthermore, mechanism‐of‐action enantiomers investigated molecular docking, defensive enzyme activity tests chlorophyll content tests. Paal–Knorr reactions are a classical method for synthesizing pyrroles, using 1,4-dicarbonyl compound and amine as reactants. It has the characteristics of green chemistry, only two molecules water lost. In this teaching learning experiment, 2,5-hexanedione 4-tert-butylaniline were used to synthesize 1-(4-(tert-butyl)phenyl)-2,5-dimethyl-1H-pyrrole without solvent catalyst. This offers several advantages, including commercially available starting materials, mild reaction conditions, solvent- catalyst-free, short time, simple convenient operation, good yield, high atom economy. The experiment allows beginners master experimental skills recrystallization, filtration, NMR, well concept chemistry. Green chemistry is important environmental protection, resource conservation, sustainable development planet. Therefore, also significant undergraduates. We present a novel approach for the skeletal rearrangement of an oxazole into azepine and pyrrole through dynamic electrocyclization process, showing innovative, unconventional reaction sequence.
