Wastewater based epidemiology has become a widely used tool for monitoring trends of concentrations different pathogens, most notably and widespread SARS-CoV-2. Therefore, in 2022, also Rhineland-Palatinate, the Ministry Science Health included 16 wastewater treatment sites surveillance program providing biweekly samples. However, mere viral load data is subject to strong fluctuations limited value political deciders on its own. state Rhineland-Palatinate commissioned University Medical Center at Johannes Gutenberg Mainz conduct representative cohort study called SentiSurv, which an increasing number up 12,000 participants have been using sensitive antigen self-tests once or twice week test themselves SARS-CoV-2 report their status. This puts fortunate position having time series both, prevalence population. Our main contribution calibration from 2023-01-08 until 2023-10-01 where we identified scaling factor ( 0.208±0.031 ) delay 5.07±2.30 days) between virus wastewater, normalized by pepper mild mottle (PMMoV), recorded SentiSurv study. The relation established fitting epidemiological model both series. We show how that can be estimate when no longer available use it as forecasting instrument several weeks ahead time. quality resulting factors depend strongly samples are normalized.

Abstract In Rhineland-Palatinate, Germany, a system of three data sources has been established to track the Covid-19 pandemic. These are number Covid-19-related hospitalizations, genecopies in wastewater, and prevalence derived from cohort study. This paper presents an extensive comparison these parameters. It is investigated whether wastewater study can be valid surrogate parameters for hospitalizations thus serve as predictors coming waves. We observe that this possible general prevalence, while suffer too large variability make quantitative predictions by purely data-driven approach. However, able detect waves qualitative manner. Furthermore, detailed different normalization techniques provided.

Isothermal exponential amplifications have been widely used in molecular based diagnostics, but their potential target nucleic acid quantification and the mechanism not yet clearly understood. Herein, we proposed most comprehensive mathematical model to date describe of isothermal amplification. Multiple critical factors that significantly affect efficiency amplification are considered with parameters definite physical significance. A method relative a gene using was proposed, formula derived. The validated via series designed experiments on loop-mediated (LAMP), which provided solid theoretical basis for According this established fundamental principal amplification, optimization strategies similar technologies given: enhance reaction by increasing enzyme activity, optimizing primer design temperature, accelerate shortening length (size) amplicon. In addition, feasibility applying LAMP-based wastewater-based epidemiology (WBE) verified experimentally.
