The invasive tomato leaf miner, Phthorimaea absoluta, is conventionally controlled through chemical insecticides. However, the rise of insecticide resistance has necessitated sustainable and eco-friendly alternatives. Entomopathogenic fungi (EPF) have shown potential due to their ability overcome minimal impact on non-target organisms. Despite this potential, precise physiological mechanisms by which EPF acts insect pests remain poorly understood. To attain a comprehensive understanding complex processes that drive successful control P. absoluta adults EPF, we investigated impacts different Metarhizium anisopliae isolates (ICIPE 665, ICIPE 20, 18) pest's survival, cellular immune responses, gut microbiota under varying temperatures. study unveiled 18 caused highest mortality rate among moths, while 20 exhibited significant reduction in total hemocyte counts after 10 days at 25°C. Moreover, both elicited notable shifts absoluta's microbiota. Our findings revealed compromised defense functions, demonstrating as biocontrol agents against production systems.

Abstract Tuta absoluta (the tomato pinworm) is an invasive insect pest with a highly damaging effect on tomatoes causing between 80 and 100% yield losses if left uncontrolled. Resistance to chemical pesticides have been reported in some T. populations. Insect microbiome plays important role the behavior, physiology, survivability of their host. In bid explore develop alternative control method, associated this was studied. study, we unraveled bacterial biota larvae adults by sequencing analyzing 16S rRNA V3-V4 gene regions using Illumina NovaSeq PE250. Out 2,092,015 amplicon sequence variants (ASVs) recovered from 30 samples (15 15 adults), 1,268,810 823,205 ASVs were obtained adults, respectively. A total 433 genera shared larval while 264 139 unique Amplicon metagenomic analyses sequences showed dominance phylum Proteobacteria adult Firmicutes dominated samples. Linear discriminant analysis size (LEfSe) comparison revealed Pseudomonas, Delftia Ralstonia be differentially enriched Enterococcus , Enterobacter Lactococcus Klebsiella Wiessella abundant larvae. Insect life processes and reproductive behaviors are significantly affected by extremely high temperatures. This study focused on Tuta absoluta, which poses a severe threat to tomato cultivars. The effects of intense heat stress the growth, development, oviposition, longevity T. absoluta were investigated. investigation encompassed various developmental stages, including eggs, pupae, adults. revealed that egg hatching pupa emergence rates reduced at temperature 44 °C maintained for 6 h. adults emerged after pupal stages exposed h was compared control. Notably, there no significant variation in adult fecundity egg-stage exposure However, all treatments exhibited control temperatures during stage. Adult survival 40 3 74.29% 22.40%, respectively, dramatically less than control, 100%. differences noted terms production. The remarkable biological and evolutionary adaptations of insects to plants are largely attributed the powerful chemosensory systems insects. tomato leaf miner (Tuta absoluta) is a destructive invasive pest with global distribution that poses serious threat production nightshade crops, especially tomatoes. Functional can attract or repel insect pests by releasing volatiles interact olfactory system insects, thereby reducing damage target crops. However, there limited research on interaction between T. absoluta genes functional plants. In this study, 97 members putative odorant-binding protein (OBP) family have been identified in whole genome absoluta. Phylogenetic analysis involving various Lepidopteran Dipteran species, including D. melanogaster, revealed OBP gene families present conserved clustering patterns. Furthermore, Plus-C subfamily showed extremely significant expansion. Moreover, expression levels varied significantly different developmental stages; is, highest number were expressed adult stage, followed larval fewer high abundance egg stage. On other hand, through Y-tube olfactometer, we plant—Plectranthus tomentosa—that repels Finally, screened response P. tomentosa at genomic level using RT-qPCR. These results laid good foundation for controlling further studying genes.

This study investigates the perceptions and practices of farmers in Pokhara, Nepal, regarding T. absoluta management. We aimed assess current adoption management strategies, identify influencing factors, uncover knowledge gaps among farmers. surveyed 69 randomly selected households involved using semi-structured questionnaire. survey collected data observations damage, (chemical, physical, pheromone-based), awareness integrated pest (IPM) ecosystem services. Our findings reveal that 94% observed damage their fields, predominant reliance chemical pesticides (86.96%). Awareness environmentally friendly practices, such as IPM, were notably lacking. Factors age household head, crop rotation, cropping systems, family type significantly influenced various practices. While 44.93% used pheromones, 31% recognized them effective. highlights severe Pokhara insufficiency Bridging eco-friendly approaches, pheromones biological controls, is crucial. emphasize need tailor extension services by considering sociodemographic factors promoting Ultimately, our will contribute more sustainable agricultural settings.

In this study, the impact of intercropping tomato (T) (Lycopersicon esculentum (Mill.) garlic (G) (Allium sativum L.) in three designs 4E:4G, 4E:8G, 4E:12G (row ratios) were assessed on density leafminer (TLM), Tuta absoluta (Meyrick), fruit infestations, diversity tomato-dwelling predators, parasitism rate, crop yields compared with sole crops over two growing seasons 2022 2023. Three intercrops caused a significant reduction number TLM eggs, larvae, galleries, as well percentage infested fruits to plots. The abundance primary Nesidiocoris tenuis (Reuter) Orius niger (Wolff), higher Furthermore, annual predators enhanced than Besides, there was noticeable enhancement Shannon index (the values 1.98 4T: 4G 2.31 12G 2023) 1.68 1.82 2023 land equivalent ratio (LER) showed notable improvement, ranging from 1.19 1.32 intercrop, crops.
