To investigate alterations in circulating microRNAs (miRNAs) the development of experimental autoimmune uveoretinitis (EAU) rats. Lewis rats were immunized with interphotoreceptor retinoid binding protein (IRBP) peptide (R14) and EAU clinical scores assessed on day 0 (baseline), days 7, 14, 21 after immunization. Total RNA was isolated from serum at same timepoints used for microarray analysis. The score peaked 14 decreased 21. Hierarchical cluster analysis principal component (PCA) miRNA expression displayed distinctly different profiles between baseline Microarray revealed significantly increased 5 (day 7), 9 14), 10 21) miRNAs, 19 20 miRNAs compared to baseline. Of note, miRNA-146a-5p, known be involved EAU, miRNA-150-5p elevated Bioinformatics that mucin type O-glycan biosynthesis cell adhesion molecules major pathways affected during EAU. PCA showed versus IRBP Upregulation miRNA-146a-5p observed effector resolution phases Analysis may help delineate systemic epigenetic changes occurring lead new insights our understanding human uveitis.
