The role of oxidants and antioxidants in inflammatory bowel disease (IBD) has been actively explored since the early 1980s, starting with respiratory burst neutrophils ischemia pathology. Since that time, enzymatic components contributing to pool reactive oxygen species, including superoxide, H2O2, lipid hydroperoxides, counteracting antioxidants—catalase, glutathione peroxidases (Gpx), peroxiredoxins (PRDX), superoxide dismutases, others—have fleshed out. My perspective on IBD is from balance or imbalance oxidant sources process. I will present evidence involvement antioxidant processes based, as much possible, my experiences Gpxs. This be discussed terms both immune system local systems. As Gpxs are generally selenium-dependent, possible deficiencies selenium uptake active impact Gpx expression explored. more recently introduced ferroptosis, an iron-dependent peroxidation-based pathological process, reviewed for its IBD.

Acute pancreatitis (AP) is a highly fatal pancreatic inflammation. In recent years, synthetic nanoparticles have been extensively developed as drug carriers to address the challenges of systemic adverse reactions and lack specificity in delivery. However, systemically administered nanoparticle therapy rapidly cleared from circulation by mononuclear phagocyte system (MPS), leading suboptimal concentrations inflamed tissues pharmacokinetics. To this challenge, we herein demonstrate surface masking strategy that involves coating selenylated Poria cocos polysaccharide with layer macrophage plasma membrane circumvent MPS sequestration, thereby enhancing therapeutic efficacy nanoparticles. Nanoparticles encapsulated membranes can simulate active homing macrophages lesions during AP, resulting excessive infiltration inflammation sites prolonged tissue retention time. This technique converts non-adhesive lipid into bioadhesive nanoparticles, increasing local accumulation under inflammatory conditions, including pancreas vulnerable lungs. The mechanism related targeting pro-inflammatory macrophages. murine models mild severe intravenous treatment macrophage-mimicking effectively reduces level diminishes recruitment neutrophils. Mechanistic studies elucidate membrane-biomimetic primarily mitigate inhibiting AKT/mTOR pathway reverse autophagic flux impairment. allows us envision biomimetic nanotherapy approach could potentially serve novel for therapy.
