Background: Tendon rupture repair can result from fibrotic scar formation through imbalanced ECM deposition during remodeling. The tissue inhibitors of matrix metalloprotease (TIMPs) not only decrease degradation, regulated by metalloproteases (MMPs), but also restrict TGF-β1 activation and thus diminish fibrosis. Methods: Rabbit tenocytes (rbTenocytes) rabbit adipose-derived stem cells (rbASCs) were cultivated under different TIMP-1 concentrations. Proliferation gene expression assessed. was incorporated into emulsion electrospun DegraPol® (DP) tubes that characterized SEM for fiber thickness, pore size, wall thickness. Static dynamic water contact angles, FTIR spectra, release kinetics determined. Results: While the proliferation rbTenocytes rbACS affected supplementation in vitro, Col1A1 increased rbTenocytes, ki67 both cell types, tenomodulin types at 100 ng/mL TIMP-1, alkaline phosphatase ALP rose significantly rbASCs. Electrospun TIMP-1/DP fibers had a ~5 μm diameter, ~10 mesh thickness ~200 μm. meshes more hydrophilic than pure DP meshes. released with sustained up to 7 days. Conclusions: may be used modulate reaction when applied around conventionally sutured tendon ruptures.
