Phenylphenalenone is an important phytoalexin for banana plant protection, yet the mechanisms governing its biosynthesis and regulation remain unclear in plant. WRKY transcription factors play essential roles modulating growth, development, of secondary metabolites. In this study, we identified 158 genes ( MlWRKYs ) from a phenylphenalenone-rich species Musella lasiocarpa . Phylogenetic analysis classified MlWRKY into three distinct subfamilies: type I, II, III. Chromosomal distribution revealed that are clustered on nine respective chromosomes. Additionally, synteny between M. Musa balbisiana uncovered highly conserved collinear regions. MIWRKY15 , MIWRKY111 MIWRKY122 were as candidate regulating PhPNs by integration multi-omics approaches. We further investigated expression pattern genes, well their putative target MlOMT22 MlOMT27 known phenylphenalenone various tissues, including leaves, stems, roots, seeds. MlWRKY15 showed similar patterns across tissues. MlWRKY122 also displayed consistent patterns, suggesting may regulate MlWRKY111 ’s was inversely correlated with indicating potential negative This study provides valuable insights family will serve useful genetic resource elucidating regulatory biosynthesis.
