Alzheimer's disease (AD) is the leading cause of dementia, and no effective treatment has been developed for it thus far. Recently, use natural compounds in neurodegenerative diseases garnered significant attention owing to their minimal adverse reactions. Accordingly, potential therapeutic effect pterostilbene (PTS) on AD demonstrated multiple vivo vitro experiments. In this study, we systematically reviewed summarized results these studies investigating PTS treating AD. Analysis literature revealed that may play a role through various mechanisms, including anti-oxidative damage, anti-neuroinflammation, anti-apoptosis, cholinesterase activity inhibition, attenuation β-amyloid deposition, tau protein hyperphosphorylation. Moreover, interferes with progression by regulating activities peroxisome proliferator-activated receptor alpha, monoamine oxidase B, Sirtuin-1, phosphodiesterase 4A. Furthermore, further elucidate mechanisms AD, employed network pharmacology molecular docking technology perform related proteins, obtained binding energies ranged from −2.83 kJ/mol −5.14 kJ/mol, indicating proteins exhibit good ability PTS. Network analysis action summary, collating summarizing relevant anticipated will serve as reference precise targeted prevention either using or other drug interventions.

Neurodegenerative diseases, encompassing conditions such as Alzheimer's disease, Parkinson's multiple sclerosis, amyotrophic lateral prion and Huntington's present a growing health concern human life expectancy increases. Despite this, effective treatments to halt disease progression remain elusive due various factors, including challenges in drug delivery across physiological barriers like the blood-brain barrier patient compliance issues leading treatment discontinuation. In response, innovative approaches leveraging noninvasive techniques with higher are emerging promising alternatives. This Review aims synthesize current options encountered managing neurodegenerative while also exploring modalities. Specifically, strategies intranasal administration nanosized systems gaining prominence for their potential enhance efficacy adherence. Nanosized systems, liposomes, polymeric micelles, nanoparticles, evaluated within context of outstanding studies. The advantages disadvantages these discussed, providing insights into therapeutic limitations. Through this comprehensive examination, contributes ongoing discourse surrounding development diseases.

Due to PTN's poor solubility and bioavailability, pterostilbene nano-emulsion gel (PTN-NEG) formulation (0.1% 0.2% w/w) was developed enhance its efficacy.
