Abstract
Background
The
highly
metastatic
triple-negative
breast
cancer
(TNBC)
relies
on
its
tumor
microenvironment
(TME)
to
maintain
phenotypic
heterogeneity
and
progression.
Extracellular
vesicles
from
hypoxic
TNBC
were
previously
shown
aid
tumoral
invasion
but
their
function
in
the
is
still
unclear.
Methods
We
used
a
novel
method
investigate
TME
vitro
called
multicellular
circulating
co-culture,
characterize
how
TNBC-derived
EVs
(EVh)
interfere
with
endothelial
cells,
fibroblasts,
monocytes
macrophages.
Results
EVh
promoted
monocyte
differentiation
M2-like
macrophages
inhibited
macrophage-derived
phagocytosis
cells.
protection
of
endothelial,
stromal
cellular
integrity
by
increased
pro-tumoral
pro-angiogenic
signaling,

fibroblasts.
Conclusion
Our
findings
highlighted
critical
role
protecting
indicating
cooperation
towards
protective
TME,
which
was
demonstrated
co-culture
conventional
protocols,
leading
an
adequate
system
for
investigating
other
tumor-related
processes,
including
cells
metastasis.
