Background:: Streptococcus equi ssp. zooepidemicus (SEZ) is one important pathogen. There are still sporadic outbreaks in China, northern United States and the Netherlands. Adenylosuccinate synthetase PurA, a newly discovered protein prior research, requires further assessment of its protective effectiveness. Methods:: In this study, we focused on expression recombinant PurA from SEZ ATCC 35246. We evaluated immunoreactivity using convalescent minipig sera. Additionally, conducted experiments mice to assess immunogenic properties. Results:: Our findings revealed that triggered substantial antibody response mice, resulting an 80% protection rate against infection. Notably, immunized with exhibited significantly reduced bacterial colonization all organs compared PBS control group. Furthermore, levels IL-6, IL-8, IL-1β, TNF-α mouse serum were elevated PurA-immunized group Hyperimmune sera targeting effectively eliminated bactericidal tests. Remarkably, antibodies demonstrated significant inhibitory effect developing biofilm. Conclusion:: Immunization elicited robust humoral cellular immune responses mice. These promising results suggest potential utility vaccine immunogens, providing valuable avenue for research into infection prevention control.
