Lipophosphoglycan (LPG) is a macromolecule on the surface of Leishmania spp. parasites. The biochemical structure LPG varies throughout parasites' life cycle between proliferative (procyclic) and infective (metacyclic) stages, as well species strains. Here, we outline protocol for growing parasites in vitro to harvest LPG. We describe steps parasite differentiation extraction purification. has applications medical research, such trained immunity immunotherapy cancer. For complete details use execution this protocol, please refer dos Santos et al.1.

Abstract Cutaneous Leishmaniasis (CL) is a significant health issue, particularly in Iraq, with notable increase cases during the winter months. This cross-sectional study was conducted at Ramadi Teaching Hospital, selected due to rising incidence of CL. A total 10 patients, aged 18 40, an average age 29, participated study, which ran from December 10, 2023, March 2024. Blood samples were collected lesions CL patients using capillary tubes, ensuring sufficient volume by multiple tubes per sample. Patients undergoing treatment Pentostam as prescribed. The blood transferred containing sodium chloride solution and stored -20°C until DNA extraction. After storage, genomic successfully extracted samples. served template for polymerase chain reaction (PCR) analysis, utilizing specific primers designed amplify genes associated virulence factors. Among samples, 7 tested positive extraction, while 3 negative. Of PCR electrophoresis indicated that GP63 gene 4 Conversely, LPG2 gene, control group included facilitate comparison both genes. Analysis confirmed Leishmania major sole causative agent Anbar.

In the mammalian hosts, Leishmania parasites survive and proliferate within phagolysosomes of macrophages. To avoid being killed by immune cells, utilize their molecules to manipulate macrophages' functions for survival. Targets such immunomodulatory are not limited macrophages, as Leishmania-derived sometimes show influence on other cells neutrophils, dendritic T B cells. This review covers research immunomodulation host immunity introduces some examples parasite-derived participating in immunomodulation. For example, cell surface lipophosphoglycan (LPG) can modulate TLR2 signaling PI3K/Akt axis macrophages leading induction Th2 Because chronic secretion inflammatory cytokines is one causes immune-mediated diseases atherosclerosis, Crohn's disease, rheumatoid arthritis, LPG may be useful a drug suppress conditions. The unique characteristics leishmanial pose promise source drugs controlling than leishmaniasis.
