Systemic sclerosis (SSc) is a systemic disease characterized by autoimmune responses, vasculopathy and tissue fibrosis. The pathogenic mechanisms involve wide range of cells soluble factors. complexity interactions leads to heterogeneous clinical features in terms the extent, severity, rate progression skin fibrosis internal organ involvement. Available disease-modifying drugs have only modest effects on halting may be associated with significant side effects. Therefore, cellular therapies been developed aiming at restoration immunologic self-tolerance order provide durable remissions or foster regeneration. Currently, SSc recommended as ‘standard indication’ for autologous hematopoietic stem cell transplantation European Society Blood Marrow Transplantation. This review provides an overview SSc, from pre-clinical models applications, opening towards more advanced therapies, such mesenchymal cells, regulatory T potentially CAR-T-cell therapies.

Background . Rheumatoid arthritis (RA) is a long-term autoimmune disease characterized by systemic immuno-inflammatory reaction leading to joint edema, synovial hyperplasia, damage cartilage and bone tissue. There currently no effective treatment for this disease. The aim of the literature review To analyze mechanisms described date that underlie effects mesenchymal stem cells (MSC) extracellular vesicles (EV) on pathogenetic links rheumatoid arthritis. Materials methods PubMed eLibrary databases period 2011–2023 were analyzed using keywords: arthritis, hematopoietic cells, factor, vesicles, cell therapy. Results discussion describes current aspects etiology pathogenesis its potential biomarkers known therapeutic strategies. effect MSC T B lymphocytes, as well other cellular participants in disease, has been studied experimental clinical studies. expands understanding postnatal progenitor their which, terms comparison, reveals advantages disadvantages each method. It can be concluded EV are promising direction however, further studies interactions molecules affecting RA needed. Additional based pathogenetically ranked approach also necessary, which allows identify patterns method with subsequent recommendation choosing use or, mainly, secretome personalized therapy, more detailed study dosage, time administration necessary.

Corneal alkali burns can cause persistent inflammation and corneal neovascularization. In this study, we divided burned rabbits into the untreated group, blank lens radiation-treated umbilical cord mesenchymal stem cells (UCMSC) UCMSC I.V. then measured inflammation, neovascularization injury repair via slit lamp microscopy, captured anterior segment optical coherence tomography (AS-OCT), performed hematoxylin-eosin staining. Compared with those in other experimental groups, lenses significantly decreased inflammatory index (IF) scores, areas of blood vessels epithelial injury. The expression interleukin (IL)-17 corneas treated was lower than that lenses, exhibited greater IL-4 signal transducer activator transcription 1 (STAT1), while cluster differentiation-3G (CD3G), a linker for activation T (LAT), IL-6, IL-1B, CC chemokine receptor 6 (CCR6) IL-23 opposite effects (all P < 0.05). Our findings demonstrated irradiated UCMSC-coated high oxygen-permeable hydrogel on ocular surface inhibited angiogenesis after alkaline burns. downregulation Th17 cell differentiation might be responsible these effects.

neuroanesthesia and critical care for neurosurgical patients, the anesthesiologist plan should have a judicious regimen of fluid management because it will be obliging in improve surgical outcomes. When fluids is replaced by good regimen, help neurosurgeon to avoid brain edema formation intravenous congestion whereas enhancing patient's volume status maintain both adequate cerebral perfusion oxygenation. We searched current available literature RCTs on MEDLINE, EMBASE Cochrane Evidence Based Medicine Reviews. This study investigates whether bone marrow mesenchymal stem cell (BMSC)-derived extracellular vesicles (EVs) can affect rheumatoid arthritis (RA) by delivering microRNA (miR)-378a-5p to regulate the interferon regulatory factor 1/signal transducer and transcription 1 (IRF1/STAT1) axis. We identified RA-associated miRNAs using GEO microarray dataset GSE121894. found most important in RA synovial tissues RT-qPCR. BMSC-derived EVs were ultracentrifuged cocultured with human microvascular endothelial cells (HSMECs) vitro. Dual-luciferase RNA immunoprecipitation studies examined miR-378a-5p’s specific binding IRF1. also measured angiogenesis, migration, proliferation CCK-8, Transwell, tube formation assays. Collagen-induced (CIA) mice models created inducing scoring it. had low miR-378a-5p expression, whereas high levels. The transfer of HSMECs boosted proliferation, angiogenesis. inhibited MiR-378a-5p-containing decreased STAT1 phosphorylation HSMEC IRF1 expression. mimic promoted dexmedetomidine phosphorylation. In CIA mice, containing enhanced vascular remodeling histopathology. Thus, from promotes inactivating IRF1/STAT1 axis preventing RA.
