The pleiotropic effects of high-density lipoprotein (HDL), including its protective properties against sepsis, are attributed to the sphingosine 1-phosphate and apolipoprotein M (ApoM) that carried on lipoproteins. In this study, we attempted elucidate possible mechanisms underlying sepsis coagulopathic state by considering modulation NETosis. Our results revealed in a lipopolysaccharide-induced mouse model, levels NETosis markers, such as plasma DNA histone, were elevated ApoM-knockout (KO) mice attenuated ApoM-overexpressing mice. ApoM-KO mice, survival rate decreased occurrence rates coagulopathy organ injury increased following administration histone. Treatment with conditioned medium cells observed HL-60S differentiated into neutrophils inhibited through suppression S1P1 or S1P4. attenuation PKCδ PKCα/β S1P4 activation may also be involved. injuries an injection histone; these partially S1P1, 3, S1P4, antagonists. Neutrophils release decondensed nuclear chromatin or Neutrophil Extracellular Traps (NETs) in response to a great number of physiological stimuli protect the host from pathogens. However, NETs have recently been shown play an important role pathogenesis autoimmune, infl ammatory, and malignant diseases. Therefore, understanding molecular mechanisms underlying formation, usually leading neutrophil death (NETosis), is extremely control aberrant chromatin. Mitogen-activated protein kinases (MAP-kinases) are involved various cellular functions such as oxidative burst, chemotaxis, degranulation, adhesion, apoptosis, but their NETosis not well understood. Three families MAP-kinases, p38, ERK1/2, JNK, described human neutrophils, we investigated contribution kinase B Akt1/2 burst using inhibitory analysis. We that MAPkinase p38 activated upon stimulation with calcium ionophore ionomycin. these induced by diacylglycerol mimetic phorbol 12-myristate 13-acetate (PMA) PMA-induced NETosis.

Oxidative burst assessed luminol-amplifi ed chemiluminescence method, NET formation immunofl uorescence analysis. shows for fi rst time that do not PMA, but A23187, a low level oxidative A23187 chemoattractant N-formyl-methionylleucyl-phenylalanine.

BACKGROUND: Ovarian cancer is the most aggressive gynecological tumor with high mortality. The ambivalent, both helper and suppressive effects of neutrophils on cells innate adaptive immunity determines their important immunoregulatory role in development malignant tumors. AIM: Evaluation effect serum interleukin-1β circulating CD16+ progression-free time advanced ovarian cancer. MATERIAL AND METHODS: In 42 primary patients serous adenocarcinoma (median age 54 years) ascites 15 benign tumors 60 admitted for examination treatment to Regional Clinical Oncology Dispensary before receiving neoadjuvant antitumor (2015–2020), level blood was determined using enzyme immunoassay, number by fluorescence microscopy monoclonal antibodies.
