Atopic dermatitis (AD) is a common skin disease worldwide. The major causes of AD are barrier defects, immune dysfunction, and oxidative stress. In this study, we investigated the anti-oxidation anti-inflammation effects Coffea arabica extract (CAE) its regulation functions in AD. vitro experiments revealed that CAE decreased reactive oxygen species levels inhibited translocation nuclear factor-κB (NF-κB), further reducing secretion interleukin (IL)-1β IL-6 induced by interferon-γ (IFN-γ)/tumor necrosis factor-α (TNF-α). Moreover, IFN-γ/TNF-α-induced NLR family pyrin domain-containing 3 (NLRP3), caspase-1, high-mobility group box 1 (HMGB1), receptor for advanced glycation end products (RAGE) expression levels. It also restored protein function-related markers including filaggrin claudin-1. vivo not only reduced redness backs mice caused 2,4-dinitrochlorobenzene (DNCB) but pro-inflammatory factors their skin. transepidermal water loss (TEWL) cell infiltration DNCB-treated mice. Overall, exerted ameliorated suggesting potential as an active ingredient treatment.

Shen chan decoction (SCD) as a significant Traditional Chinese medicine (TCM) to treat atopic dermatitis (AD), but its mechanism of action has not been clarified, so we started the present study, first possible effects SCD on AD were predicted using network pharmacology. Next, dinitrochlorobenzene was used establish mouse model AD. After successful modelling, administered intragastrically mice. Eventually, KEGG pathway enrichment analysis indicated that improved mainly through inflammation and gut microbiota. The experimental findings revealed treatment attenuated symptoms downregulate characteristic immune factors, namely IL-4, IL-6 IgE. Moreover, it promoted balance between Th1/Th2 cells. Furthermore, itch signaling pathways involving H1R/PAR-2/TRPV1 inhibited. 16S rRNA sequencing results administration influenced Firmicutes/Bacteroidetes ratio at phylum level by augmenting relative proportions Lactobacillaceae Muribaculaceae family genus levels, while decreasing abundances Lactococcus Ruminococcus. These suggest internal is an effective therapeutic approach for We may be alternative therapy AD.Additionally, could offer valuable insights into pathogenesis development innovative agents.

(1) Background: Atopic dermatitis (AD) is characterized as a chronic inflammatory skin disease with significant incidence rate. The pathophysiological mechanisms underlying AD remain incompletely understood. However, extensive research demonstrates that complex interplay among genetic, immune, and environmental factors contributes to the disruption of barrier function. Inflammation identified one pathological in AD. Recombined mussel adhesive protein exhibits anti-inflammatory properties. recombinant has been used less frequently for AD, so we explored therapeutic effect potential mechanism. (2) Methods: We established mice model vivo an LPS-induced inflammation HaCaT cells vitro. Through assessment lesion scores, itch frequency, transepidermal water loss, microcirculation, HE staining, Elisa assays IL-6, IL-12, IL-13, IL-4, IL-5, IFN-γ, IgE, TNF-α, immunohistochemical staining filaggrin CK14, Masson Western blot analysis NF-κB p65, P-P65, Keap1, Nrf2, effects recombined on symptoms, pathology, inflammation, its are investigated. (3) Results: significantly improved compromised barrier, reduced scratching frequency mice, decreased lowered expression factors, thus ameliorating damage. Mechanistically, downregulated P-p65/p65 Keap1 while upregulating level Nrf2. (4) Conclusions: Overall, our results demonstrate effectiveness attenuating DNFB-induced by inhibiting activating Keap1/Nrf2 signaling pathway. Thus, promising candidate treatment
