Esculin and esculetin are 2 widely studied coumarin components of Cortex Fraxini , which is a well-known herbal medicine with 2000-year history. In vivo in vitro studies have demonstrated that both variety pharmacological activities, including antioxidant, anti-tumor, anti-inflammatory, antibacterial, antidiabetic, immunomodulatory, anti-atherosclerotic, so on. Their underlying mechanisms action biological activities include scavenging free radicals, modulating the nuclear factor erythroid 2-related pathway, regulating cell cycle, inhibiting tumor proliferation migration, promoting mitochondrial pathway apoptosis, NF-κB MAPK signaling pathways, CD4 + T cells differentiation associated cytokine release, vascular smooth muscle cells, etc. This review aims to provide comprehensive information on esculin esculetin, noteworthy importance exploring therapeutic potential compounds.

Abstract Pneumonia is a serious and life‐threatening lung inflammation with high morbidity mortality. Accumulating evidence has suggested that esculin, derivative of coumarin, possesses potent anti‐inflammatory effects. This study designed to explore the pharma role underlying mechanism esculin against lipopolysaccharides (LPS)‐induced pneumonia. TC‐1 cells were stimulated by LPS mimic inflammatory injury model in vitro . Cell viability, proliferation, apoptosis determined using MTT assay, 5‐ethynyl‐2′‐deoxyuridine flow cytometry. Background/Objectives: Acute lung injury (ALI) is an inflammatory disorder affecting patients in intensive care with high mortality. No specific pharmacological treatment available. Musa paradisiaca L. (banana) a cosmopolitan plant, and homemade syrup from its inflorescence used many countries to treat pulmonary inflammation. Therefore, this study analyzed the hydroalcoholic extract (HEM) of on ALI experimental model. Methods: Swiss mice were challenged lipopolysaccharide treated HEM after 1, 24, 48 h (five animals/group, three times). Results: The HEM-treated presented decrease neutrophil migration bronchoalveolar lavage fluid (BALF), alveolar region, blood, correlating downregulation CD18 expression. also reduced protein concentration BALF, caused edema formation, impaired NF-κB activation via inhibition TLR4 signaling pathway, decreased IL-1β, TNF-α production, free DNA release, myeloperoxidase (MPO) activity. However, induced increased IL-10 BALF. Conclusions: HEM’s anti-inflammatory immunomodulatory activities are by deactivating neutrophils decreasing receptor, MPO activity inducing production.
