Hepatocellular Carcinoma (HCC) features a complex pathophysiology and unpredictable immunosuppressive microenvironment, which limit the effectiveness of traditional therapies lead to poor patient outcomes. Understanding immune characteristics HCC is essential for elucidating microenvironment developing more effective treatments. This study investigates role Peptidyl-prolyl isomerase H (PPIH) in by analyzing its expression, prognosis, methylation levels, relationship with cell infiltration. We utilized bulk sequencing clinical data from UCSC Xena GTEx database preprocessing subsequent differential expression analysis PPIH tumor adjacent normal tissues, evaluating prognostic parameters like overall survival disease-free interval between low high groups. Immune infiltration was analyzed via CIBERSORT ssGSEA, while DNA somatic mutation analyses were performed using MExpress "maftools", respectively, alongside vitro vivo experiments assess PPIH's functional roles. Our findings indicated that significantly upregulated various cancer types, correlating increased mutations, altered gene patterns. High levels linked enhanced T regulatory (Treg) decline Th17 populations, impacting vital pathways related damage repair proliferation. Furthermore, knockdown led reduced viability, proliferation, invasion promoting apoptosis. In vivo, repressed growth modified attenuating potentially increasing Treg accumulation. emphasizes critical progression facilitating modulating landscape, thereby positioning as potential therapeutic target management.

Laryngeal squamous cell carcinoma (LSCC) is a highly aggressive malignancy with rising incidence over time. The tumor microenvironment (TME) plays crucial role in LSCC development, yet the precise cellular characteristics of laryngeal cancer and its TME remain unclear. Here, we employed single-cell RNA sequencing analysis to uncover heterogeneous populations immune cells investigate LSCC. This revealed significant heterogeneity among malignant cells, T macrophages. Notably, regulatory were markedly increased at sites, macrophage identified an presence Macrophage-C1-C1QC subset up-regulated PPIF expression. Bulk RNA-seq further confirmed up-regulation exosomes derived from tissues. Consistently, survival indicated that high expression was associated poor prognosis Further analyses suggested enhancement their anti-inflammatory phenotype promotion F11R-F11R signaling allowing evade macrophage-mediated cytotoxicity. Our study provides new insights into dynamics highlights critical progression, offering potential therapeutic targets for treatment.

Abstract Prostate cancer is a malignant tumor of the male urological system with highest incidence rate in world, which seriously threatens life and health middle-aged elderly men. The progression prostate involves interaction between cells microenvironment. Understanding mechanisms pathogenesis disease important to guide diagnosis therapy. emergence single-cell RNA sequencing (scRNA-seq) spatial transcriptome (ST-seq) technologies has brought breakthroughs study cancer. It makes up for defects traditional techniques such as fluorescence-activated cell sorting that are difficult elucidate cell-specific gene expression. This review summarized heterogeneity functional changes microenvironment revealed by scRNA-seq ST-seq, aims provide reference optimal treatment
