Acquired
drug
resistance
is

failure
and
high
mortality,
highlighting
the
necessity

novel
intervention
targets.
Circular
RNAs
(circRNAs),
an
abundant
class
of
RNA
molecules
with
closed
loop
structure,
possess
characteristics
including
stability,
which
provide
unique
advantages
in
clinical
application.
Growing
evidence
indicates
that
aberrantly
expressed
circRNAs
are
associated
against
various
treatments,
targeted
therapy,
chemotherapy,
radiotherapy,
immunotherapy.
Therefore,
targeting
these
aberrant
may
offer
strategy
improve
efficiency
therapy.
Herein,
we
present

their
regulatory
roles
on

ABSTRACT
The
incidence
of
colorectal
cancer
(CRC)
has
been
increasing
in
recent
decades.
Current
methods
for
CRC
screening
have
their
own
drawbacks,
thus
there
is
an
urgent
need
to
identify
the
key
microbes
that
drive
development
wider
application
early
detection
and
prevention
CRC.
To
address
this
issue,
we
performed
fecal
microbiome
analysis
by
high-throughput
sequencing
16S
rRNA
gene
combined
with
blood
biochemical
indicators
patients
stages
I,
II,
III,
IV,
healthy
people,
polyps.
Fecal
microbiota
was
disturbed,
as
evidenced
significantly
reduced
α-diversity
stage
IV
markedly
different
β-diversity.
random
forest
model
identified
top
25
genera
from
174
training
data,
resulting
a
diagnostic
accuracy
87.95%.
Further,
combining
differential
analysis,
screened
out
11
biomarkers
changed
groups.
Peptostreptococcus
,
Parvimonas
Shewanella
Oscillibacter
Eggerthella
Gemella
associated
were
enriched,
while
Fenollaria
Staphylococcus
Ezakiella
Finegoldia,
Neisseria
remission
suppressed

carcinoembryonic

microbial
biomarkers,
carbohydrate
19-9
(CA
19-9)

co-occurrence

genus
level

reshape
microenvironment
(TME)
through
metabolite
secretion,
modulation
of
immune
pathways,
direct
interactions
with
host
cells,
thereby
affecting
biology
outcomes.
Despite
substantial
heterogeneity
among
types,
recent
insights
underscore
microbiome’s
potential
as
both
diagnostic/prognostic
biomarker
targetable
component
for
innovative
treatments.
In
this
review,
we
synthesize
emerging
knowledge
mechanistic
roles
tumor-associated
microbiota
in
shaping
TME,
focus
how
these
discoveries
can
guide
novel
strategies.
We
further
explore
interdisciplinary
advances,
including
convergence

nanotechnology,
to
enhance
drug
delivery,
