Abstract Chemical modifications such as acetylation and carboxymethylation significantly influence the physicochemical properties of biopolymers, including exopolysaccharides (EPS). In this study, glucan EPS from Enterococcus hirae OL616073 was chemically modified to acetylated (AG) carboxymethylated (CG) via an affordable process. The structure were investigated by 1H NMR spectroscopy, Fourier transform infrared (FTIR), scanning electron microscope (SEM), contact angle, particle size, thermal analysis (TGA-DSC), X-ray diffractometer (XRD). results demonstrated effect modification on its properties. As L* varied between 70.13 74.46 with a mild darkening in derivative than carboxymethylated, a* ranged 3.55–3.87, affirming reddish tint all powders. There slight increase (70.24) whiteness index (WI) for CG comparison AG (68.10) Untreated (UT) (65.23). Decrease bulk density (0.532 0.415 g/mL), tapped (0.756 0.531g/mL), Hausner ratio (1.42 1.27), compressibility (CI) (29.61 21.81%) UT observed. Micrographs revealed significant change agglomerated granules AC rough collapsed CG. Water activity (aw) untreated decreased 0.358 0.345 further increased 0.380 size (226.6 nm) 365.8 nm 526.4 Acetylation enhanced hydrophobicity (water angle: 56.58° 66.28°), while became more hydrophilic (44.13°). holding capacity 328.11% 494.22% (CG). emulsion stability retained better at different time intervals. Furthermore, showed antitumor effects against HCT116 cells (8.18 47.04% reduction) (2.04 17.22%) 25–500 µg/mL. findings highlight functional groups reaction conditions properties, suggesting their potential food development.
