Abstract Point mutations in the acetylcholinesterase-1 gene (ace-1) have been associated with resistance to OPs many insects. However, presence and function of ace-1 OP Helicoverpa armigera (Lepidoptera: Noctuidae), a significant lepidopteran pest damaging wide range crops, remain largely unexplored. This study investigated insecticide phoxim 12 field populations H. from northern China 2022, revealing low levels (2.5- 6.7-fold). Using an amplicon sequencing approach, we screened for 13,874 moths collected 114 between 2006 2022. We found 3 amino acid substitutions (A201S, G227E, F290V) potentially related resistance. The mean frequencies A201S, F290V were 0.0032, 0.0001, respectively. To assess these mutations’ role resistance, expressed wild-type mutant AChE1 proteins Sf9 cells. Biochemical characterization revealed 3.1-fold 3.3-fold increase I50 chlorpyrifos-oxon A201S mutants compared enzyme, correlating 2.9-fold 2.7-fold decrease Ki value. Genetically modified crops that produce insecticidal proteins from Bacillus thuringiensis (Bt) are currently the most efficient and safest method of pest control worldwide. However, prolonged planting period has led to a reduction in efficacy Bt due evolution resistance field. This review paper examines status lepidopteran pests under field conditions, elucidates molecular mechanism underlying their Cry toxins, discusses management strategies based on these mechanisms. Abstract Cotton plays a crucial role in shaping Indian economy and rural livelihoods. The cotton crop is prone to numerous insect pests, necessitating insecticidal application, which increases production costs. advent of the expression Bacillus thuringiensis (Bt) protein has significantly reduced burden pest without compromising environmental or human health. After introduction transgenic cotton, cultivated area expanded 22 million hectares, with 64% increase adoption by farmers worldwide. Currently, Bt accounts for 93% India.
