Ashwagandha (Withania somnifera) is a well-known adaptogenic herb traditionally used to enhance sleep quality and mitigate stress-induced cognitive decline. This study investigated the effects of different doses ashwagandha root extract (AE) formulations on function, oxidative stress, neuronal plasticity in rat model deprivation (SD). Forty-nine rats were randomly assigned seven groups: control, wide platform (WP), SD, SD + A1 (15 mg/kg AE 1.5%), A2 (30 A3 (5.5 8.0%), A4 (11 8.0%). The was administered orally for four weeks. induced via modified significantly impaired spatial memory, increased suppressed GABA receptor activity. Treatment with all doses, except 15 1.5%, considerably reduced serum corticosterone (12% A2, 15% A3, 32% A4), CRH (11% 14% 17% ACTH (22% 26% 38% MDA levels (31% 34% 46% A4) (p < 0.05). All improved antioxidant enzyme activity memory performance, while 8.0% notably serotonin (19% 33% dopamine (40% 50% A4). Moreover, treatment enhanced markers partially GABAergic function. These findings suggest that formulations, particularly at higher concentrations, exert neuroprotective against SD-induced impairment by modulating neurotransmitter balance, neuroplasticity, indicating their potential application managing stress-related neurological disorders.
