Purpose of review Chronic kidney disease-mineral bone disorder (CKD-MBD) is associated with several adverse outcomes, including fragility and sarcopenia. Identification new agents mitigating systemic damage related to uremia critical needed unveil pathways implicated in CKD-MBD. Recent findings Osteopontin (OPN) involved different physiological pathological processes works as a bridge connecting systems. It may serve biomarker for many diseases, human cancers, neurodegenerative disorders autoimmune diseases. OPN has been disturbances mineralization remodeling, an interplay parathyroid hormone FGF23 experimental models. In patients CKD severe hyperparathyroidism, expression muscle tissue linked worse functionality local inflammation, which partially reverted after parathyroidectomy. Summary Future studies could confirm the role nephrology. Greater understanding its CKD-MBD will help us define better therapeutic strategy CKD.

Extracellular matrix remodeling is a natural response to injury but, excessive extracellular accumulation, or fibrosis, causative factor in hundreds of diseases that limit organ function, regenerative responses, and can interfere with therapies. Fibrosis closely related inflammation, both which occur the salivary glands patients treated radiation for head neck cancers suffering from autoimmune conditions, such as Sjögren's Disease. Despite known involvement fibrosis disease inhibitory effects on tissue regeneration, mechanisms through elaborated gland are poorly understood. Stromal fibroblasts primary matrix-producing cells drive inflammation. To define temporal responses injury, we induced temporary obstructive though ligation submandibular sublingual ducts then performed single-cell RNA sequencing pathway analysis at timepoints immediately following injury. Using bioinformatic approaches, identified three unique fibroblast groups dynamically respond We characterized changes matrisomal inflammatory gene expression over 7-day time course one group be injury-responsive fibrogenic cell type. Understanding how early later timepoints, along defining signaling pathways regulated by fibroblasts, could lead better understanding contribution acute facilitate development therapeutics minimize promote chronic states.

Urinary obstruction causes injury to the renal papilla and leads defects in ability concentrate urine which predisposes progressive kidney injury. However, regenerative capacity of after reversal is poorly understood. To address this, we developed a mouse model reversible urinary characterized by extensive papillary injury, followed robust regeneration response complete histological recovery over 3- month period. these mice have pronounced defect concentrating capacity. We now show that this due permanent changes composition, organization, transcriptional signatures epithelial, endothelial, interstitial cell lineages papilla. There are persistent inflammatory responses also seen patients with stone disease but associated cell-specific adaptive increasingly hypoxic environment obstruction. Taken together, our analysis new reveals partial repair structure function all major cellular compartments include both shared distinct different types humans mice.
