Antigenic peptide-based mRNA vaccines have been explored for immunotherapeutic use in various types of cancer because their advantages activating durable and specific immune responses. However, role modulating tumor metastasis is still unclear. Here, we identify a conserved linear epitope-based peptide, Ma3P, located the proteolytic region major histocompatibility complex (MHC) class I-related chain A (MICA) α3 further design mCM10-L, an vaccine that encodes carrier protein CRM197 10 tandem repeats Ma3P. We demonstrate vaccination with mCM10-L induces production antibodies block MICA/B α1/2 shedding, activate CD8+ T cells natural killer (NK) cells, significantly inhibit MICA/B+ mice. Furthermore, stimulation triggers to promote MICA/B-mediated killing in-vitro-interacting human organoid model humanized Our results indicate potential clinical application prospects vaccine.

Melanoma is a highly invasive malignancy with poor prognoses in advanced stages. Developing risk model that can accurately assess prognosis and guide personalized treatment crucial for improving the clinical management of melanoma. This study aims to develop validate an immune-based prognostic melanoma through comprehensive bioinformatics analysis. We collected transcriptomic data from multiple public databases identified 9 immune features significantly associated using single-sample Gene Set Enrichment Analysis (ssGSEA) Cox regression. These were utilized construct model, which was subsequently validated relevant bulk datasets single-cell GEO database, encompassing diverse patient populations sample types. The effectively stratified patients into high-risk low-risk groups distinct survival outcomes. Further analysis revealed significant associations between genomic heterogeneity indicators, such as tumor mutational burden (TMB), loss heterozygosity (LOH), checkpoint gene expression. robustness confirmed data, highlighting key genes potential therapeutic relevance.
