Abstract In 2020–2021, a “mysterious illness” struck Senegalese fishermen, causing severe acute dermatitis in over one thousand individuals following exposure through drift-net fishing activity. Here, by performing deep analysis of the environmental samples we reveal presence marine dinoflagellate Vulcanodinium rugosum and its associated cyclic imine toxins. Specifically, show that toxin PortimineA, strongly enriched samples, impedes ribosome function human keratinocytes, which subsequently activates stress kinases ZAKα P38 promotes nucleation NLRP1 inflammasome, leading to release IL-1β/IL-18 pro-inflammatory cytokines cell death. Furthermore, cell-based models highlight naturally occurring mutations P38-targeted sites are unable respond PortimineA exposure. Finally, development use organotypic skins zebrafish demonstrate ZAKα-NLRP1 axis drives skin necrosis inflammation. Our results exemplify threats health caused emerging toxins identify NRLP1 as important pharmacological targets mitigate toxicity.

Inflammasomes are multiprotein complexes of the innate immune system that mediate inflammatory responses to infection and local systemic stress tissue injury. Sepsis is defined as a condition related to infection that manifests with multiorgan dysfunction, representing life-threatening state. Consequently, severe complications frequently occur, liver injury being one of the most prevalent serious sepsis. Liver dysfunction during sepsis serves an independent predictor mortality. This review provides comprehensive overview current research on sepsis-induced (SILI), encompassing clinical manifestations, diagnostic criteria, pathogenesis and therapeutic strategies associated this condition. SILI may manifest hypoxic hepatitis due ischemia shock, cholestasis resulting from abnormal bile metabolism, or duct sclerosis. The pathophysiology involves intricate interactions among inflammatory response, oxidative stress, cell death. All these factors complicate treatment represent potential targets for intervention. Furthermore, addresses limitations inherent in conventional therapies currently employed managing emphasizes novel targeted aimed at addressing fundamental mechanisms underlying

Viruses are obligate intracellular pathogens as their replication depends on the metabolism of host cell. The induction cellular suicide, known programmed cell death (PCD), has potential to hinder viral and act a first line defense against pathogens. Apoptosis, necroptosis, pyroptosis three important PCD modalities. Different signaling pathways involved in execution, they also differ ability cause inflammation. Cytomegaloviruses (CMV), beta-herpesviruses with large double-stranded DNA genomes, encode great variety immune evasion genes, including several suppressors. While CMV inhibitors apoptosis necroptosis have been studied for years, inhibitor identified characterized only recently. Here, we describe how human murine interfere apoptosis, pathways. We discuss importance different forms containment spread vivo.

Previous studies by our group and others have highlighted the critical role of hyperinflammation in pathogenicity duck hepatitis A virus 1 (DHAV-1), an avian picornavirus that has caused significant devastation industry worldwide for decades. However, precise mechanisms which DHAV-1 infection regulates inflammatory responses, particularly production IL-1β, remain poorly understood. In this study, we demonstrate triggers NF-κB- NLRP3 inflammasome-mediated IL-1β production. Mechanistically, infection, its replication translation, disrupts cellular homeostasis Ca
