This study aims to assess the associations of complete blood cell count (CBC)-derived inflammatory biomarkers with prevalence asthma and mortality.Data was collected from 1999-2018 National Health Nutrition Examination Survey (NHANES). Mortality identified using Death Index until December 31, 2019. The analyzed relationship between CBC-derived biomarkers, including neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte (PLR), monocyte-to-lymphocyte (MLR), systemic response index (SIRI), immune-inflammation (SII), multiple logistic regressions. To significance in predicting all-cause respiratory disease mortality patients, Cox proportional regressions random survival forest (RSF) analysis were utilized.A total 48,305 participants included, a mean age 47.27 ± 0.18 years 49.44% male. Asthma and atopic dermatitis (AD) represent significant global health challenges in children. This study aimed to investigate trends incidence, prevalence, disability-adjusted life years (DALYs) for childhood asthma AD from 1990 2021. The utilized information the Global Burden of Disease (GBD), Injuries, Risk Factors Study sample size this consisted children with or between ages 0 14. From 1990–2021, we calculated AD's age-standardized DALYs by area, age, sex, socio-demographic index. In 2021, prevalence reached 95.7 million cases (age-standardized rate: 4,758 per 100,000), Low SDI region recording 25.4 cases. For AD, was 72.4 3,600 predominantly Middle regions (19.7 cases). Between incidence rates decreased both conditions. These molecularly targeted agents provide significant benefits for patients unresponsive to conventional treatments by addressing underlying immune mechanisms, particularly type 2 inflammation driven cytokines such as IL-4, IL-5, IL-13. Recent advancements include biologics targeting alarmins like thymic stromal lymphopoietin (TSLP) IL-33, which may address both non-type inflammation, broadening their therapeutic scope. Despite effectiveness, remain expensive, posing socioeconomic challenges, there are concerns regarding long-term safety inter-individual variability in responses. Abstract Background The role of lung group 2 innate lymphoid cell (ILC2) activation in allergic asthma is increasingly established. However, the regulatory mechanisms underlying hypoxia‐inducible factor‐1α (HIF‐1α)‐mediated glycolysis ILC2‐mediated airway inflammation remain unclear. Objective To investigate HIF‐1α/glycolysis axis inflammation. Methods Glycolysis and HIF‐1α inhibitors were used to identify their effect on function glucose metabolism mouse human ILC2s vivo vitro. Blocking mice under interleukin‐33 (IL‐33) stimulation performed test ILC2 responses. Conditional HIF‐1α‐deficient confirm specific ILC2‐driven models.
