This study aimed to detect the causal association of overweight and obesity on severe COVID-19 infection with respiratory failure through a two-sample Mendelian randomization (MR) method based genome-wide studies datasets. All summary data exposures outcome used in this were obtained from IEU database derived Europeans. The mainly inverse variance weighted test relationship. Simultaneously, MR-PRESSO MR-EGGER pleiotropy, sensitivity analysis was performed using leave-one-out analysis. In analyses, we found no between (e.g., OR = 1.15, 95% CIs 0.96-1.37, P .13 for obesity-ebi-a-GCST90000255), subtypes 1.93, 0.90-4.14, .10 other hyperalimentation) as well (OR 0.90, 0.64-1.27, .54) failure. findings showed or Further validation is needed regarding whether risk factor it.

The human gut microbiome, composed of a vast array microorganisms that have co-evolved with humans, is crucial for the development and function brain systems. Research has consistently shown bidirectional communication between through neuronal, endocrine, immunological, chemical pathways. Recent neuroscience studies linked changes in microbiome microbial metabolites to various neuropsychiatric disorders such as autism, depression, anxiety, schizophrenia, eating disorders, neurocognitive disorders. Novel metagenome-wide association confirmed these variations large samples expanded our understanding interactions genes microbiome. causal relationship microbiota being elucidated establishment cohort incorporating data advanced statistical techniques. Ongoing animal focused on microbiota-gut-brain axis are promising developing new prevention treatment strategies conditions. scope broadened from microbiome-modulating therapies including prebiotics, probiotics, synbiotics postbiotics more extensive approaches fecal transplantation. systematic reviews meta-analyses strengthened evidence base innovative treatments. Despite research over past decade, many intriguing aspects still need be regarding role therapeutic interventions

Previous studies have reported an association between gut microbiota and cirrhosis. However, the causality intestinal flora liver cirrhosis still remains unclear. In this study, bi-directional Mendelian randomization (MR) analysis was used to ascertain potential causal effect microbes cirrhosis.Large-scale Genome Wide Association Study (GWAS) data of were obtained from FinnGen, Mibiogen consortium, a GWAS meta-analysis Alcoholic (ALC). Two-sample MR performed determine relationship Furthermore, employed examine direction relations. In analysis, we found that 21 microbiotas potentially associated with reverse 11 displayed associations genetic liability in microbiome We family Lachnospiraceae (OR: 1.59, 95% CI:1.10-2.29) might be harmful cirrhotic conditions (ICD-10: K74). genus Erysipelatoclostridium protective factor for (OR:0.55, CI:0.34-0.88) PBC (OR:0.68, CI:0.52-0.89). Combining results firstly identified Genus Butyricicoccus had on (Forward: OR: 0.37, CI:0.15-0.93; Reverse: 1.03, CI:1.00-1.05).We new provided insights into role pathological progression

Background There is growing evidence of associations between the gut microbiota and anxiety disorders, where changes in microbiotas may affect brain function behavior via microbiota-gut-brain axis. However, population-level studies offering a higher level for causality are lacking. Our aim was to investigate specific associated metabolites that closely related disorders provide mechanistic insights novel management perspectives disorders. Method This study used summary-level data from publicly available Genome-Wide Association Studies (GWAS) 119 bacterial genera phenotype “All disorders” reveal causal effects on identify with A two-sample, bidirectional Mendelian randomization (MR) design deployed, followed by comprehensive sensitivity analyses validate robustness results. We further conducted multivariable MR (MVMR) analysis potential impact neurotransmitter-associated metabolites, bacteria-associated dietary patterns, drug use or alcohol consumption, lifestyle factors such as smoking physical activity observed associations. Results Bidirectional identified three causally disorders: genus Eubacterium nodatum group Ruminococcaceae UCG011 were protective, while an increased risk Further MVMR suggested metabolite-dependent mechanism, primarily driven tryptophan, tyrosine, phenylalanine, glycine cortisol, which consistent previous research findings, probably played significant role mediating these Furthermore, modifying pattern salt, sugar processed meat intake, adjusting state levels, appears be effective approaches targeting manage Conclusion findings offer avenues developing precise metabolites.
