Artemisia argyi, a significant medicinal plant in China, is known for its high content of essential oils, flavonoids, and other bioactive compounds. MYB transcription factors are the largest gene family plants widely reported to play important roles development, metabolism, defense, stress resistance. However, A. argyi has not been systematically studied. The aim this study was comprehensively analyze explore potential role flavonoid biosynthesis. Here, phylogeny, chromosome location, structure, cis-acting elements, expression patterns Gene ontology (GO) annotation members were investigated using bioinformatics methods based on whole-genome transcriptome data argyi. In total, 227 AYMYB identified from genome, including 22 1R-MYB, 165 R2R3-MYB, 16 3R-MYB, 5 4R-MYB 19 atypical members. These AYMYBs unevenly distributed across genome. Subcellular localization prediction revealed that all localized nucleus. protein motifs, conserved domains, structures identified, results showed same subfamily exhibited similar motifs structures. Cis-acting elements GO analysis suggested may be involved many biological processes related Moreover, quantitative real-time PCR (qRT-PCR) approximately 50 genes levels leaves specific under MeJA treatment. Together, our research will offer useful information future investigations into functions especially regulating process biosynthesis response
