Immune and metabolic factors play an important role in the onset development of insomnia. This study aimed to investigate causal relationship between insomnia immune cells metabolites. Data for 731 cell phenotypes, 1400 metabolites, this were obtained from GWAS open-access database. Two-way Mendelian randomization was used (1) detect (2) identify potential mediating analysis identified eight phenotypes with a insomnia, two protective namely CD8br %T CD80 on CD62L + myeloid dendritic cells. The other six risk i.e., CD4/CD8br, CD16-CD56 NKT, CCR2 cells, CD40 monocytes, CD38 CD3-CD19-, CD25 CD45RA CD4 not Treg. Further revealed 11 metabolites that causally related Five 3-hydroxy-3-methylglutarate, cholate, dodecanedioate, N-formylmethionine, x-26054. Six 3-amino-2-piperidone, 6-oxopiperdine-2-carboxylate, caffeine theophylline ratio, leucine, maltose, x-24736. In addition, our showed leucine mediated association CD4/CD8br From genetic information, we confirmed eleven metabolite levels. Notably, found leucine-mediated providing evidence supporting plasma serving as mediators.

Doxorubicin-induced myocardial injury, characterized by hypertrophy and heart failure (HF), represents a primary contributor to end-stage cardiovascular mortality associated with anthracycline drugs. Prior research has elucidated that SIRT6-mediated oxidative processes mitochondrial metabolic reprogramming are pivotal in sustaining energy metabolism during damage cardiomyocytes. In the aftermath of doxorubicin-induced fibrosis exacerbate impairment cardiac ejection function, resulting elevated oxygen consumption. This condition is accompanied disrupted ATP production, diminished biogenesis, inadequate synthesis new DNA, collectively triggering necroptosis apoptosis pathways. Our preliminary experimental results have confirmed SIRT6, traditional medicine, exerts cardioprotective effects. Nevertheless, interaction between SIRT6 Nrf-2-mediated biogenesis context HF remains inadequately understood. The generation mitochondria key mechanism involved DNA repair cell cycle management.

Myocardial fibrosis is a common manifestation of end-stage cardiovascular disease, but there lack means to reverse fibrosis. Astragaloside IV (AS-IV), the major active component Astragalus membranaceus Fisch. ex Bunge Fabaceae, possesses diverse biological activities that have beneficial effects against disease. This systematic review aims summarize anti-fibrosis effect AS-IV in animal models (rats or mice only) and its underlying mechanisms, provide potential directions for clinical use AS-IV. PubMed, EMBASE, Web Science, CNKI, Wanfang database, SinoMed were searched from inception 31 December 2024. The following characteristics included studies extracted summarized: model, route administration, dose/concentration, measurement indicators, mechanisms. quality was assessed used 10-item scale SYRCLE. represents promising multi-target candidate myocardial treatment 24 eligible analysis. first comprehensively evaluate mechanisms across heterogeneous disease models, including infarction, hypertension, ischemia-reperfusion injury, myocarditis. may include collagen metabolism, anti-apoptosis, anti-inflammation and, pyroptosis, antioxidants, improving mitochondrial function, regulating senescence, etc. Current evidence remains preclinical, with critical gaps toxicological profiles, human safety thresholds, adverse reaction data. Future research must integrate robust evaluations, optimized combination therapies, adaptive trials validate translational potential.
