Regulatory T cells (Tregs) are a specialized subset of suppressive that essential for maintaining self-tolerance, regulating effector cells, managing microbial infections, preventing tumors, allergies, and autoimmune disorders, facilitating allograft transplantation. Disruptions in Treg function or abundance contribute to an imbalance between pathogenic protective immune diseases. Recently, one promising treatment strategy restore balance involves the selective expansion manipulation Tregs using low-dose IL-2 therapy, adoptive cell transfer, chimeric antigen receptor (CAR)-Treg approaches. have been shown increasing number research studies prevent even treat variety such as allergic diseases, transplant rejection, graft-versus-host disease. A thorough comprehension is anticipated provide clear prospects effective immunotherapy wide range This review provides overview biology, including their functions, mechanisms, phenotypic markers, well involvement disease settings. Furthermore, we discuss therapeutic potential different subpopulations translational applications

Diabetes mellitus (DM) refers to a group of chronic diseases with global prevalence, characterized by persistent hyperglycemia resulting from various etiologies. DM can harm organ systems and lead acute or complications, which severely endanger human well-being. Traditional treatment mainly involves controlling blood sugar levels through replacement therapy drugs insulin; however, some patients still find satisfactory curative effect difficult achieve. Extensive research has demonstrated close correlation between enteric dysbacteriosis the pathogenesis types DM, paving way for novel therapeutic approaches targeting gut microbiota manage DM. Fecal transplantation (FMT), method re-establishing intestinal microbiome balance, offers new possibilities treating diabetes. This article provides comprehensive review microbiota, as well current advancements in FMT using an illustrative example. study aims offer perspectives establish theoretical foundation clinical diagnosis management

The gut microbiota offers numerous benefits to the human body, including promotion of nutrient absorption, participation in metabolic processes, and enhancement immune function. Recent studies have introduced concept gut-organ axis, which encompasses interactions such as gut-brain gut-liver gut-lung axis. This underscores complex interplay between various organs tissues, brain, heart, lungs, liver, kidneys, muscles, bones. Growing evidence indicates that can influence onset progression multi-organ system diseases through their effects on Traditional Chinese medicine has demonstrated significant efficacy regulating gastrointestinal system, leveraging its unique advantages. Considerable advancements been made understanding role axis within mechanisms action traditional medicine. review aims elucidate roles health, explore potential connections microbiota, examine therapeutic microbiota-gut-organ Furthermore, addresses limitations challenges present current research while proposing directions for future investigations this area.

Autoimmune rheumatic diseases (ARDs) are a heterogeneous group of conditions characterized by excessive and misdirected immune responses against the body's own musculoskeletal tissues. Their exact aetiology remains unclear, with genetic, demographic, behavioural environmental factors implicated in disease onset. One prominent hypothesis for initial breach tolerance (leading to autoimmunity) is molecular mimicry, which describes structural or sequence similarities between human microbial proteins (mimotopes). This similarity can lead cross-reactive antibodies T-cell receptors, resulting an response autoantigens. Both commensal microbes microbiome pathogens trigger thereby potentially contributing onset ARDs. In this review, we focus on role mimicry rheumatoid arthritis systemic lupus erythematosus. Moreover, implications also briefly discussed ankylosing spondylitis, sclerosis myositis.

Atrial cardiomyopathy is a multifaceted heart disease characterized by structural and functional abnormalities of the atria closely associated with atrial fibrillation its complications. Its etiology involves number factors, including genetic, infectious, immunologic, metabolic factors. Recent research has highlighted critical role gut microbiota in pathogenesis cardiomyopathy, this consistent gut–heart axis having major implications for cardiac health. The aim work to bridge knowledge gap regarding interactions between particular focus on elucidating mechanisms which dysbiosis may induce remodeling dysfunction. This article provides an overview changes composition effects metabolites. We also discuss how diet exercise affect influencing microbiota, as well possible future therapeutic approaches targeting axis. A healthy can prevent disease, but ecological lead variety symptoms, induction disease. pathophysiological aspects impact structure function, strategies exploring modulation treatment cardiomyopathy. Fecal microbiota transplantation (FMT) is the most effective therapy for preventing recurrent Clostridioides difficile infection (rCDI). However, impact of FMT formulations and storage conditions on bacterial viability, community structure, functionality, clinical efficacy remains under-investigated. We studied effect different viability (live/dead staining cell sorting), structure (16S rDNA analysis), metabolic functionality (fermentation) frozen lyophilized formulations. The success rates rCDI patients were correlated retrospectively with formulations, durations, host factors using Edmonton program database. Bacterial remained at 10–20% across various was comparable to that fresh FMT. Live dead fractions in both preparations exhibited distinct structures. Storage but not temperatures, negatively affected diversity. More short-chain fatty acids found metabolomic profiling vitro fermentation products than Clinical 537 receiving a single dose significantly among three longer durations advanced recipient age impacted efficacy. Together, our findings suggest should be considered when establishing guidelines product shelf life optimal treatment outcomes.
