Pseudomonas aeruginosa (P. aeruginosa) causing urinary tract infections (UTIs) are a major concern among hospital-acquired infections. The need for an effective vaccine that reduces the is imperative. This study aims to evaluate efficacy of multi-epitope encapsulated in silk fibroin nanoparticles (SFNPs) against P. aeruginosa-mediated UTIs. A constructed from nine proteins using immunoinformatic analysis, expressed, and purified BL21 (DE3) cells. encapsulation efficiency SFNPs 85% with mean particle size 130 nm 24% antigen released after 35 days. formulations adjuvanted or alum significantly improve systemic mucosal humoral responses cytokine profile (IFN-γ, IL-4, IL-17) mice. Additionally, longevity IgG response maintained at least 110 days steady state. In bladder challenge, mice treated admixed demonstrate significant protection kidneys aeruginosa. highlights promising therapeutic potential
